In an assay for determination of beta-lactam antibiotic resistance in a target bacterial strain, the strain is grown in the presence of both a beta-lactamese inducing antibiotic and a beta-lactam indicator antibiotic [which kills or inhibits the growth of bacteria] unable to hydrolyze beta-lactam antibiotics. Growth, indicative of drug resistance in the target strain, is monitored by detecting a fluorophor released by the enzymatic cleavage of a metabolizable fluorogenic compound.

The present invention relates to a method to determine the concentration of an unknown substance in a colorimetric reaction using a fluorometric reader. The change in color can be monitored by observing the change of fluorescence of a known amount of a suitable fluorophore in the reaction chamber. Under appropriate conditions the absorption spectrum of the chromophore overlaps with the emission spectrum of the fluorophore thereby allowing the change in fluorescence to be proportional to the intensity of color in the reaction and thus, proportional to the quantity of the substance of interest. A multi-layer blood culture sensor includes two matrices. The first matrix is a polymer that is permeable to carbon dioxide and water, but impermeable to protons. A pH sensitive absorbance based dye is encapsulated or isolated in the polymer. The second matrix is a polymer with a pH insensitive fluorescent dye encapsulated or isolated in the polymer. The matrices are spectrally coupled and are useful for the determination of microorganisms in a blood culture bottle.

In particular, a change in color can be monitored by observing the measurement of fluorescence intensity of a fluorophore in an inert matrix. The absorption spectrum of the chromophore may overlap the excitation and/or emission spectrum of the fluorophore, thereby allowing the change in fluorescence to be related to the intensity of color in the reaction and thus related to the quantity of the substance of interest.
