A microfluidic device includes multiple microfluidic assay units arranged on a substrate, with each assay unit including multiple scaffold regions each containing a three-dimensional scaffold with associated cells, and a media channel surrounding a fluid-permeable boundary portion of at least a second scaffold region, wherein a fluid-permeable interface between the media channel and the second scaffold region comprises a curved shape spanning an arc of more than 90 degrees. A third scaffold region may be provided. Boundaries between different scaffold regions, and between a scaffold region and the media channel, may include microposts that may be spaced apart in a curved configuration. A method for performing an assay utilizing such a device is further provided.

Microfluidic devices and methods incorporating assay units with multiple 3D scaffold regions

A scaffold-free microtissue is disclosed that includes one or more gold nanostructures linked to a functional moiety, wherein the functional moiety is one or more vasculogenic peptides, one or more anti-inflammatory peptides, one or more antiapoptotic peptides, one or more antinecrotic peptides, one or more antioxidant peptides, one or more oligonucleotides, one or more lipid particles, one or more phospholipid particles, one or more liposomes, one or more nanoliposomes, one or more microRNAs, or one or more siRNAs. The scaffold-free microtissue further includes a plurality of cardiac myocytes or cardiac myoblasts, which are conjugated to the one or more gold nanostructures, wherein the plurality of cardiac myocytes or cardiac myoblasts are arranged in a cluster. The scaffold-free microtissue further includes a plurality of fibroblasts, wherein the fibroblasts are arranged in at least one layer of fibroblasts that substantially surrounds the cluster of gold-nanostructure-conjugated cardiac myocytes or gold-nanostructure-conjugated cardiac myoblasts.

Nano scale decoration of scaffold-free microtissue using functionalised gold nanostructures

A microfluidic device for more accurately modeling the in vitro environment in which cancer occurs is disclosed. The microfluidic device includes a surface defining one or more microfluidic channels, a first three dimensional scaffold comprising one or more cancer cells that is spatially separated from the one or more microfluidic channels, a second three dimensional scaffold, at least a portion of which is contacting and in fluid communication with the first three dimensional scaffold, and that is spatially separated from the one or more microfluidic channels, and a third three dimensional scaffold, at least a portion of which is contacting and in fluid communication with the one or more microfluidic channels and the second three dimensional scaffold. The device can be used to assay anti-cancer agents, or as a system for modeling the growth, behavior, or metastasis and tumor formation of cancer cells.

The present invention provides a temperature-responsive dual-gelling hydrogel comprising a plurality of hydrogel polymers and a polymer cross-linking moiety, wherein the LCST of the hydrogel polymers is less than 37&#xb0; C., and the polymer cross-linking moiety is capable of chemically cross linking to the hydrogel polymers to form a polymer matrix.

Injectable cell-laden biohybrid hydrogels for cardiac regeneration and related applications

The device comprises a surface defining one or more microfluidic channels that may further comprise one or more endothelial cells, a first three dimensional scaffold comprising one or more cancer cells that is spatially separated from the one or more microfluidic channels, and a second three dimensional scaffold comprising one or more stromal cells, at least a portion of which is interposed between the one or more microfluidic channels and the first three dimensional scaffold. The second three dimensional scaffold is in fluid communication with both the first three dimensional scaffold and the one or more microfluidic channels. The device can be used to assay anti-cancer agents, or as a system for modeling the growth, behavior, or metastasis and tumor formation of cancer cells.
