An automated radiosynthesizer device includes a plurality of reactor assemblies, each reactor assembly being operatively connected to a horizontal actuator for moving the reactor assembly in the horizontal direction and a vertical actuator for moving the reactor assembly in the vertical direction. A plurality of disposable cassettes are disposed above each of the plurality of reactor assemblies, each cassette comprising a lower surface comprising a plurality of sealed and un-sealed gaskets, wherein the un-sealed gaskets are connected to internal fluid paths within the cassette. The device includes a three-axis reagent and gas handling robot disposed above the plurality of cassettes and terminates in a vial gripper and a gas manifold having an inert gas port and a vacuum port. The device includes a control system configured to control the horizontal actuator and vertical actuator of each reactor assembly and the three-axis reagent and gas handling robot.

Automated, multi-pot high-pressure radio-synthesizer for production of pet tracers

The automated synthesis of clinically relevant amounts of 16&#x3b2;-F-fluoro-5&#x3b1;-dihydrotestosterone (F-FDHT) using a commercially available radiosynthesizer. Synthesis was performed in 90 minutes with a decay-corrected radiochemical yield of 29&#xb1;5%. The specific activity was 4.6 Ci/&#x3bc;mol (170 GBq/&#x3bc;mol) at end of formulation with a starting activity of 1.0 Ci (37 GBq). The formulatedF-FDHT yielded sufficient activity for multiple patient doses and passed all quality control tests required for routine clinical use.

Inter alia, the first titania-catalyzed [F]-radiofluorination in highly aqueous medium is provided. In embodiments, the method utilizes titanium dioxide, 1:1 acetonitrile-thexyl alcohol solvent mixture and tetrabutylammonium bicarbonate as a base. Radiolabeling may be directly performed with aqueous [F]fluoride without the need for drying/azeotroping step, which reduces radiosynthesis time while keeping high fluoride conversion. The general applicability of the synthetic strategy to the synthesis of the wide range of PET probes from tosylated precursors is demonstrated.
