The present disclosure relates to a modified Fc fragment, antibodies containing the same and the use thereof. The Fc fragment is derived from human IgG1, and the constant region CH2 domain of the Fc fragment contains multiple substitutions. Such substitution can significantly reduce the binding ability of Fc fragment to Fcγ receptors (FcγR), and reduce the non-specific activation of T cells by antibodies (such as anti-CD3 antibodies).

Modified Fc fragment, antibody comprising same, and application thereof

Provided in the present application is a humanized and multifunctional CD3 antibody; a heavy chain variable region of the CD3 antibody comprised herein contains the amino acid sequence of any one of SEQ ID NOs: 1-3 or a variant sequence thereof; and a light chain variable region thereof contains the amino acid sequence of any one of SEQ ID NOs: 26-28 or a variant sequence thereof. Provided are compositions and methods for preparing a cell suitable for producing a bispecific antibody. A plurality of eukaryotic cells are incubated with an agent under conditions to allow the cells to arrest at G1/S phase. The agent is then removed from the cells and the cells are transfected with a first vector comprising a sequence encoding a first monovalent antigen-binding unit having specificity to a first antigen and a second vector comprising a sequence encoding a second monovalent antigen-binding unit having specificity to a second antigen. A cell is identified from the plurality of cells that expresses both the first and the second antigen-binding units.

Preparation and selection of cells for producing bispecific antibodies

The present invention provides a bispecific antibody. The bispecific antibody provided by the present invention comprises a single-chain unit and a monovalent unit, wherein the single-chain unit has a specific binding capability against a surface antigen CD3 of an immune cell; the monovalent unit has a specific binding capability against a surface antigen HER2 of a tumor cell; the single-chain unit comprises a single-chain variable fragment ScFv fused with an Fc fragment; and the monovalent unit comprises a light chain and heavy chain pair. The present invention also provides a preparation method of the bispecific antibody and pharmaceutical use of these antibodies.
