This invention concerns a family of chimeric antibodies with high affinities to a high molecular weight, tumor-associated sialylated glycoprotein antigen (TAG-72) of human origin. These antibodies have (1) high affinity animal V,and V,sequences which mediate TAG-72 binding and (2) human C,and C,regions. They are thought to produce significantly fewer side-effects when administered to human patients by virtue of their human C,and C,antibody domains. The nucleotide and amino acid sequences of V,&agr;TAG V,, CC46 V,, CC49,, CC83 V,, and CC92 V,, and CC49,, CC83 V,, and CC92 V,idiotype sequences are disclosed, as well as in vivo methods of treatment and diagnostic assay using these chimeric antibodies.

The present invention discloses novel chimeric monoclonal antibodies directed against human carcinoembryonic antigen, having antigen-specific variable regions. DNA constructs for the light and heavy chain variable regions comprising the novel antibodies of the invention are also disclosed. Family of anti-carcinoembryonic antigen chimeric antibodies

A novel class of polypeptides of the general formula (F-(Pro).sub.n).sub.m F, wherein F represents a flexible amino acid sequence wherein each amino acid is individually selected from the group consisting of serine, glycine, and threonine, and n is an integer from 4-8 inclusive and m is an integer from 1-4 inclusive, is disclosed. Thses polypeptides are useful in the construction of conjugates between antibodies and peptide toxins. The preparation of such conjugate toxins by linking antibodies to toxin/spacer composites prepared by recombinant techniques is also disclosed.

The invention concerns a method for suppressing bone marrow which comprises administering at least one bone marrow suppressing radionuclide composition selected from the group of radionuclide compositions comprised of at least one of Samarium-153, Gadolinium-519, and Holmium-166 and at least one of ethylenediaminetetramethylenephosphonic acid, diethylenetriaminepentamethylenephosphonic acid, hydroxyethylethylenediaminetrimethylenephosphonic acid, nitrilotrimethylenephosphonic acid and tris(2-aminoethyl)aminehexamethylenephosphonic acid, or a physiologically acceptable salt thereof.
