Novel materials and methods useful for expressing heterologous proteins in prokaryotic cells are provided, including prokaryotic cells expressing FkpA and/or Skp.

Methods and materials for enhancing functional protein expression in bacteria

The expression vectors and methods using anexpression system for the large scale production of FGF18 are described. The vectors utilize the FGF18 coding sequence with specific changes in nucleotides in order to optimize codons and mRNA secondary structure for translation in. Using the expression vectors, the FGF18 gene was produced into a level of greater than 1 g/L in fed batch fermentation. Also included are OmpT deficientstrains, as well as OmpT and fhuA negative strains transformed with an FGF18 expression vector.

FGF18 production in prokaryotic hosts

Homogeneous preparations of IL-28A, IL-28B, and IL-29 have been produced by mutating one or more of the cysteine residues in the polynucleotide sequences encoding the mature proteins. The cysteine mutant proteins can be shown to either bind to their cognate receptor or exhibit biological activity. One type of biological activity that is shown is an antiviral activity.
