Abstract
The present invention provides methods for assembling DNA molecules in a predetermined order to produce a DNA construct useful inmediated transformation in plants. The method employs ligation independent cloning of separate DNA elements where one of the DNA elements contains T-DNA borders from. The invention further provides methods for assembling a construct containing an inverted repeat. Using this approach, DNA constructs are constructed rapidly, efficiently and directionally.
