Abstract

Apparatus and methods for high resolution holographic recordation and microscopic examination of the holographic image of biological tissue. A single beam Denisyuk holographic recordation of the biological tissue is obtained with direct contact or close proximity to the tissue. Contrast enhancement by staining with a dye selected to maximize absorption of the light frequency used to record the hologram is used to improve contrast and resolution. A liquid interface is formed between the recording film and the subject tissue to improve image quality and resolution. The resulting high resolution three dimensional holographic images may be examined under microscope magnification for such applications as medical biopsy.
