Abstract

The present disclosure provides a binding protein comprising: a polypeptide heavy chain comprising: VH-(X)n-VH—CH (X)y wherein VHis a first variable domain, VHis a second variable domain, CH is a constant domain, Xrepresents an amino acid or peptide, Xrepresents an Fc region, n is 0 or 1 and y is independently 1 or 2, and a polypeptide light chain comprising: VL-(X)n-VL-C wherein VL1 is a first variable domain, VLis a second variable domain, C is a constant domain, Xrepresents an amino acid or peptide and n is 0 or 1, wherein the heavy chain and light chain are aligned such that VHand VLform a first binding domain, and VHand VLform a second binding domain and wherein: there is a disulfide bond between VHand VL, and/or there is a disulfide bond between VHand VL, and use thereof in treatment.
