The present disclosure provides a nucleated red blood cell simulating particle, which may be leukocytes bound to a fluorescent-staining inhibitor capable of stably binding to the nucleus or a nucleic acid in a cell so as to reduce the binding capacity of the particles to a fluorescent dye during their detection. The present disclosure also provides a method for preparing nucleated red blood cell simulating particles, including the following steps: (a) obtaining purified leukocytes; (b) suspending the leukocytes in a cell treatment solution containing a fluorescent-staining inhibitor which stably binds to the nucleus or a nucleic acid in a cell, and (c) washing the obtained product. The present disclosure also provides a hematology control mixture containing the nucleated red blood cell simulating particles. In addition, the present disclosure describes the use of the nucleated red blood cell simulating particles and the hematology control mixtures comprising the same, for the quality control of a blood cell analyzer.

Preparation and use of nucleated red blood cell simulating particles and hematology control mixtures

In one aspect of the present disclosure there is provided a method for preparing reticulocyte mimetics. In another aspect of the present disclosure there are provided reticulocyte mimetics obtained using the method. In yet another aspect of the present disclosure there is provided a whole blood reference control including the reticulocyte mimetics as provided. In still another aspect of the present disclosure there is provided a composition useful for preparing reticulocyte mimetics.

Reticulocyte mimetics and method of preparation of the same

A method for preparing five-part differential leukocyte analogs from red blood cells includes selecting appropriate red blood cells; treating the red blood cells with a multi-functional reagent system for maintenance of a structural integrity of the membranes of the red blood cells and synchronous adjustment of cell volume, morphology and inclusions; subjecting the treated red blood cells to an intensified fixation; and washing the treated cells for preservation. The present disclosure also relates to leukocyte analogs prepared by the above method, a reagent system used for preparing the analogs, and a quality control and a calibrator including the analogs for use in a hematology analyzer.

The present invention relates to a processing method for long-term stabilization of biological red blood cell volume. The red blood cells can be further used in reference materials and calibrators of a hematology analyzer. The method of the present invention comprises contacting the sample with a solution comprising a membrane-fixation reagent and an aldehyde for a processing period to terminate or slow down the red blood cell metabolism. It is provided by experiments that such process enables the volume of the red blood cells to be stable in a period over 120 days.
