The present invention provides assays and methods for efficiently testing a polynucleotide of interest for a phenotype in a root. In some embodiments, the assays and methods include regenerating green tissue that is transgenic for at least one polynucleotide of interest into one or more transgenic plantlets that have at least one transgenic root. Further provided are methods of making a root assay by contacting green tissue with a first rooting medium to produce a plantlet and a plurality of roots. Additionally provided are methods of assaying for insecticidal activity on a live root. Accordingly provided herein is a substantially contamination-free, root bioassay. Further provided are methods of identifying a promoter having activity in a root.

In vitro plant bioassay methods

Methods for producing homozygous plants, seeds, and plant cells are provided. The methods comprise transforming a somatic cell of a maize haploid embryo with a polynucleotide of interest and treating the transformed cell with a chromosome doubling agent. One or more growth stimulation proteins, such as, for example, RepA or Lec1, may also be provided.

The invention provides improved plant transformation methods. In particular the method provides increased transformation frequency, especially in recalcitrant plants. The method includes various transformation protocols for monocots, such as maize and sorghum, using a combination of media and light conditions to achieve increased efficiency of monocot transformation and increased callus initiation frequencies.

Highly transformable elite inbred line-PHWWE

Methods for plant transformation, for improving transformation efficiency, and for producing transgenic plants are provided. The methods comprise crossing a recipient plant from a genetic line of a plant species of interest with a donor plant selected from a transformation competent genetic line of the same plant species or of another closely related plant species to obtain a hybrid plant. Tissues obtained from the hybrid plant are transformation competent. These tissues can then be transformed with one or more nucleotide sequences of interest and selected for transgenic events having the nucleotide sequence of interest integrated within a chromosome derived from the recipient plant. Transformed cells can be selected and transgenic hybrid plants regenerated. The nucleotide sequence of interest can be introgressed into the genetic line from which the original recipient parent was derived, or into other genetic lines. Transformed plants and seeds are additionally provided.

Methods of transforming plants and identifying parental origin of a chromosome in those plants

The present invention relates to methods for improving the transformation frequency of -mediated transformation of maize embryos. A preferred method for transforming maize using comprises the steps of: contacting at least one immature embryo from a maize plant with capable of transferring at least one gene to said embryo; co-cultivating the embryos with culturing the embryos in medium comprising N6 salts, an antibiotic capable of inhibiting the growth of and a selective agent to select for embryos expressing the gene; and regenerating plants expressing the gene.

Methods and compositions for the efficient transformation of sorghum is provided. The method involves infection with Agrobacterium, particularly those comprising a super-binary vector. The transformed gene will be flanked by at least one T-DNA border and present in the transformed sorghum in low copy number. Transformed sorghum, cells, tissues, plants, and seed are also provided.
