Retinal diseases, such as age-related macular degeneration (AMD), are the leading cause of blindness in the elderly population. Since no known cures are currently present, it is crucial to diagnose the condition in its early stages so that disease progression is monitored. Systems and methods for detecting and mapping the mechanical elasticity of retinal layers in the posterior eye are disclosed herein. A system including confocal shear wave acoustic radiation force optical coherence elastography (SW-ARF-OCE) is provided, wherein an ultrasound transducer and an optical scan head are co-aligned to facilitate in-vivo study of the retina. In addition, an automatic segmentation algorithm is used to isolate tissue layers and analyze the shear wave propagation within the retinal tissue to estimate mechanical stress on the retina and detect early stages of retinal diseases based on the estimated mechanical stress.

Shear wave based elasticity imaging using three-dimensional segmentation for ocular disease diagnosis

An in vivo optical biopsy applicator of the vaginal wall for treatment planning, monitoring, and imaging guided therapy is described herein. The applicator may include an imaging probe operatively coupled to a laser ablation device. The applicator allows for non-invasive optical tissue monitoring in order to define pre- and post menopausal parameters, pre- and post-treatment microscopic changes, and offers an objective scientific tool in order to compare currently available medical, non-medicated, and energy-based treatment protocols.

Optical biopsy applicators for treatment planning, monitoring, and image-guided therapy

Ciliary motion in the upper airway is the primary mechanism by which the body transports foreign particulate out of the respiratory system. The ciliary beating frequency (CBF) is often disrupted with the onset of disease. Current imaging of ciliary motion relies on microscopy and high speed cameras, which cannot be easily adapted to in-vivo imaging. M-mode optical coherence tomography (OCT) imaging is capable of visualization of ciliary activity, but the field of view is limited. The present invention features the development of a spectrally encoded interferometric microscopy (SEIM) system using a phase-resolved Doppler (PRD) algorithm to measure and map the ciliary beating frequency within an on face region. This novel high speed, high resolution system allows for visualization of both temporal and spatial ciliary motion patterns.

Mapping ciliary activity using phase resolved spectrally encoded interferometric microscopy

A device for cancer screening and triage using an optical coherence tomography (OCT) imaging system integrated with optical imaging probe is provided. Endoscopic OCT images are generated using the OCT system having a helical probe. The images are further analyzed to generate a depth resolved intensity OCT signal to classify the region of tissue into variable grades of dysplasia to guide the physician's biopsy or resection.

Optical coherence tomography for cancer screening and triage

An OCT system with a swept light source centered around 1.7 &#x3bc;m for identifying atherosclerotic plaque and visualization of large lipid pool is provided. Advantages for using the 1.7 &#x3bc;m swept source laser include higher contrast between lipid and normal tissue and deeper penetration of the optical signals from the 1.7 &#x3bc;m swept source laser into the tissue. With deeper penetration into the tissue, more structural information from deep within the tissue is obtained. The present invention also features multimodality imaging systems that integrate additional imaging systems into the OCT system at 1.7 &#x3bc;m. As an example, an integrated 1.3 &#x3bc;m and 1.7 &#x3bc;m OCT system is provided which simultaneously generates OCT images using both the OCT systems which are used to characterize and differentiate the types of tissue.

Optical coherence tomography device for characterization of atherosclerosis with a 1.7 micron swept laser source

A multimodal medical device for the diagnosis and/or thermal treatment of a disease includes an instrument for performing thermal therapy; and an imaging system having multiple tomographic imaging modes, including optical coherence tomography (OCT), ultrasound imaging, photoacoustic (PA) imaging, fluorescence imaging and/or thermal imaging to provide guidance for accurate diagnosis and treatment of the disease.

Integrated intraoperative diagnosis and thermal therapy system

A system for imaging and quantifying shear wave and shear modulus under orthogonal acoustic radiation force (ARF) excitation using the OCT Doppler variance apparatus. The ARF perpendicular or with at least a perpendicular component to the OCT beam is produced by a remote ultrasonic transducer. The OCT Doppler variance apparatus, which is sensitive to the transverse vibration, is used to measure the ARF induced vibration. For analysis of the shear modulus, the Doppler variance apparatus is utilized to visualize shear wave propagation. The propagation velocity of the shear wave is measured and then used to quantitatively map the shear modulus.

Optical coherence elastography (OCE) method under acoustic radiation force excitation using OCT doppler variance methods and OCT correlation-based methods

A three-dimensional (three dimensional) segmentation method with intensity-based Doppler variance (IBDV) based on swept-source OCT. The automatic three dimensional segmentation method is used to obtain seven surfaces of intra-retinal layers. The microvascular network of the retina, which is acquired by the IBDV method, can be divided into six layers. The microvascular network of the six individual layers are visualized, and the morphology and contrast images can be improved by using the segmentation method. This method has potential for earlier diagnosis and precise monitoring in retinal vascular diseases. Each tomographic image is composed of eight repeat scans at the same position, which achieves a high time difference to improve the sensitivity of the angiographic method. A subpixel registration algorithm is used to reduce the eye movement. The GPU was used to accelerate the data processing to achieve real-time preview of the acquired data.

Automatic three-dimensional segmentation method for OCT and doppler OCT angiography

The invention includes an improvement in a method of assessing a gemstone having at least one planar face with an internally reflecting surface including the steps of optically modifying the at least one planar face of the gemstone to return a sample beam from an internally reflecting plane corresponding to the at least one planar face to an optical coherence tomography (OCT) system; selectively directing the sample beam from an optical coherence tomography (OCT) system onto the gemstone; and generating an OCT image map of the gemstone to determine volume, gem carat weight and/or quality.

Integrated multimodality intravascular imaging system that combines optical coherence tomography, ultrasound imaging, and acoustic radiation force optical coherence elastography

Integrated ultrasound, OCT, PA and/or florescence imaging endoscope for diagnosing cancers in gastrointestinal, respiratory, and urogenital tracts

A method using optical coherence tomography to capture the microvascular network of the superficial layer of the finger skin for the purpose of fingerprint authentication and liveness detection. At the dermal papilla region, the vascular pattern follows the same pattern of the fingerprint and this vascular pattern forms a live vascular fingerprint. This live vascular fingerprint provides for ultrahigh security and a unique way for fingerprint-based personal verification. Because the system is based on blood flow, which only exists in a living person, the technique is robust against spoof attaching. After performing non-contact in-vivo imaging of a human fingertip, a three dimensional vasculature image is reconstructed from a plurality of vasculature tomography images and at least one vasculature fingerprint image which corresponds to the fingertip is extracted from the three dimensional vasculature image. This extracted image may then be compared to known fingerprint database for authentication or for liveness detection.

Apparatus and method for capturing a vital vascular fingerprint

Ultrasound guided optical coherence tomography, photoacoustic probe for biomedical imaging

A fiber-delivered probe suitable for CARS imaging of thick tissues is practical. The disclosed design is based on two advances. First, a major problem in CARS probe design is the presence of a very strong anti-Stokes component in silica delivery fibers generated through a FWM process. Without proper spectral filtering, this component affects the CARS image from the tissue sample. The illustrated embodiments of the invention efficiently suppress this spurious anti-Stokes component through the use of a separate fiber for excitation delivery and for signal detection, which allows the incorporation of dichroic optics for anti-Stokes rejection. Second, the detection of backscattered CARS radiation from the sample is optimized by using a large core multi mode fiber in the detection channel. This scheme produces high quality CARS images free of detector aperture effects. Miniaturization of this fiber-delivered probe results in a practical handheld probe for clinical CARS imaging.
