Wastewater containing organic matter may be treated using a multi-zone apparatus. In a first zone, organic matter in the wastewater may, among other things, be converted to at least volatile fatty acids (VFAs) and, thereafter, a portion of the treated wastewater may flow to a second zone that may, among other things, convert the VFAs to methane.

Bioelectrochemical Systems (BES) for use as Biological Oxygen Demand (BOD) sensors, systems incorporating BES sensors for measuring BOD, and methods of using the sensors and systems for measuring BOD. The disclosed sensors are inexpensive to construct, long-lasting, have a fast response, and a large dynamic range. The invention includes biological oxygen demand (BOD) sensors which incorporate at least three working electrodes, at least one counter electrode, a reservoir for dilution fluid, and a sensor for measuring an electric current or a voltage which flows from the working electrodes to the counter electrode. The BOD sensors will typically also include at least one electrically active microbe disposed in proximity to the working electrode.

Biological oxygen demand sensors

Disclosed are nucleoside 5'-triphosphates modified at the &#x3b1;, (&#x3b2;, and/or &#x3b3; phosphate to substitute one or more of the phosphate oxygen atoms with a borano, seleno, and/or thio group. Derivatives and pharmaceutically acceptable salts are also contemplated herein. Also disclosed are processes for preparing nucleoside 5&#x2032;-triphosphates. Non-limiting, exemplary processes can include first reacting salicyl phosphorochloridite with a pyrophosphate reagent to provide P2, P3-dioxo-P1-(salicyl) cyclotriphosphite, and then reacting P2, P3-dioxo-P1-(salicyl) cyclotriphosphite with a nucleoside, followed by oxidation of the nucleoside-phosphite intermediate and hydrolysis to form the modified nucleoside 5&#x2032;-triphosphate. Nucleoside 5&#x2032;-triphosphates can be useful in nucleic acid replication, transcription, and translation. Processes disclosed herein are not limited to nucleoside 5&#x2032;-triphosphates having a native base moiety, such as 9-adeninyl, 1-cytosinyl, 9-guaninyl, 1-thyminyl, or 1-uracilyl, and are also effective for the preparation of nucleoside 5&#x2032;-triphosphates having a modified nucleobase or unnatural nucleobase.

Synthesis of nucleoside 5'-triphosphates and their derivatives

In one aspect, the present invention provides reactor designs, component designs, and operating schemes for removing nitrates and chemical oxygen demand from any suitable wastewater stream. In another aspect, the invention also provides reactor designs, component designs, and operating schemes designed to modify and improve pH and water quality in wastewater streams.

Denitrification and pH control using bio-electrochemical systems

Disclosed is an RNA aptamer, a synthetic oligonucleotide of 2'-fluoro-modified A, U, and C nucleotides, with improved stability compared to its unmodified counterpart. Like the unmodified aptamer, however, the modified version is a potent glutamate receptor antagonist. Additionally, the RNA aptamers described herein are water soluble by nature, and generally exhibit nano- to micromolar potency making them potential therapeutic agents for the treatment of neurological disorders involving glutamate receptor activity.

RNA hydrogels

Disclosed are methods and compositions for inhibiting DNA synthesis in a cell using RNA. Inhibition of DNA synthesis by RNA can be used, for example, in analytical methods, as a research tool to affect cells under study, to synchronize cell cycle in a cell culture, and to inhibit cell growth. For example, inhibition of DNA synthesis in cancer cells can be used to inhibit cancer cells and treat cancer. The RNA can be any RNA, such as whole cell RNA, whole cell mRNA, whole cell ribosomal RNA, whole cell transfer RNA, synthetic RNA, recombinant RNA, modified RNA, or a combination. The composition can comprise RNA and a pharmaceutically acceptable carrier or RNA, a targeting molecule, and a pharmaceutically acceptable carrier. The targeting molecule can be a tumor-targeting peptide.

Methods and compositions for interference with DNA polymerase and DNA synthesis

Methods for crystallizing a molecule of interest, such as a polypeptide, in complex with nucleic acid, including contacting the molecule of interest with selenium-derivatized nucleic acid and crystallizing the molecule of interest/selenium-derivatized nucleic acid complex are provided. Methods for determining the X-ray crystal structure of molecule of interest/selenium-derivatized nucleic acid complexes are also provided. Typically, the method of X-ray crystal structural determination includes selenium single-wavelength anomalous phasing of the selenium-derivatized nucleic acid. In some embodiments the phases for the X-ray crystal structure of the molecule of interest are not provided from another crystal. Also disclosed are methods of affecting a biological process by administering a functional nucleic acid to a cell or a subject and/or by bringing into contact a nuclease and a functional nucleic acid, where the functional nucleic acid is selenium-derivatized nucleic acid.

Methods for structural determination of selenium derivatized nucleic acid complexes

The disclosure relates to synthetic oligonucleotides that are unique in that they are RNA molecules that have the capacity to form a hydrogel. Also disclosed are DNA oligonucleotides that encode the RNA oligos so that the oligos can be prepared using in vitro transcription. The disclosure further pertains to pharmaceutical compositions comprising these hydrogels.

Disclosed are compositions which can mimic DNA and/or RNA in cells of a subject and methods of using them as a substrate in testing efficacy of one or more compositions in reducing and/or preventing radiation, such as ultraviolet (UV) radiation-caused DNA and/or RNA damage of said subject. Ultraviolet and high-performance liquid chromatography methods for the evaluation of sunscreen efficacy

Inhibitors of AMPA-type glutamate ion channels are useful as biochemical probes for structure-function studies and as drug candidates for a number of neurological disorders and diseases. Disclosed herein is the identification of an RNA inhibitor or aptamer by an in vitro evolution approach and characterization of its mechanism of inhibition on the sites of interaction by equilibrium binding and on the receptor channel-opening rate by a laser-pulse photolysis technique. The aptamer of the invention is a noncompetitive inhibitor of AMPA-type glutamate ion channels, one that selectively inhibits the GluA2QAMPA receptor subunit without any effect on other AMPA receptor subunits or on kainate or NMDA receptors. Furthermore, the aptamer preferentially inhibits the closed-channel state of GluA2Qwith a K=1.5 &#x3bc;M or by &#x2dc;15-fold over the open-channel state. The potency and selectivity of this aptamer rival those of small molecule inhibitors. Together, these properties make the aptamers of the present invention promising water-soluble, highly potent, GluA2 subunit-selective drugs.
