The present invention provides synthetic rodent antibody libraries, such as mouse or rat antibody libraries, as well as polypeptides, nucleic acids, vectors, host cells and methods used in conjunction with these libraries. The present invention also provides antibodies isolated from such libraries and variants of such antibodies.

Rodent combinatorial antibody libraries

The present disclosure enables collections of variable heavy chain and variable light chain pairs comprising, in part, germline protein sequences that are pre-selected for functional properties relevant to developability, wherein the collections may be used to select against any antigen using, for example, phage display.

Collection and methods for its use

The present invention relates to methods for the formation of inter-molecular disulphide bonds, including (poly)peptides/proteins, nucleic acids, vectors, host cells and bacteriophages used in these methods. Furthermore the invention relates to the use of this method for the improved display of (poly)peptides/proteins on the surface of bacteriophage particles.

Methods for the formation of disulphide bonds

A tricistronic vector (i.e., a vector capable of expressing three exogenous genes, which are not fused together, under the control of one promoter) effectively can encode an immunoglobulin-presenting polypeptide and two immunoglobulin (Ig) polypeptides. The encoded Ig-presenting polypeptide is able to associate with at least one of the Ig polypeptides via co-expressed associating agents. A vector according to the present invention particularly is suited for phage display technology, e.g., when the Ig-presenting polypeptide is a phage coat protein and the Ig polypeptides associate to form a Fab.
