A HER2-targeting agent containing any of the antibody or antigen-binding fragment thereof, and a pharmaceutical composition containing the HER2-targeting agent are disclosed.

The present invention provides a method for producing an antibody against podocalyxin expressed specifically in cancer cells. The method includes a step of introducing a nucleic acid encoding all or a portion of podocalyxin into cells expressing a cancer cell-specific sugar chain structure to cause cancer cell-specific podocalyxin or a portion thereof to be expressed therein, a step of immunizing a non-human mammal with the cancer cell-specific podocalyxin or portion thereof to obtain antibodies, and a step of purifying the antibodies by primary screening using purified cancer cell-specific podocalyxin or a portion thereof.

Object of the present invention is to provide various anti-podoplanin antibodies useful as a drug or reagent. The present invention provides an anti-podoplanin antibody or antigen-binding fragment thereof, each having an epitope in any of the following regions in the amino acid sequence of podoplanin represented by SEQ ID NO: 1: (i) from position 56 to position 80, (ii) from position 81 to position 103, (iii) from position 81 to position 88, and (iv) from position 25 to position 57.
