A milk-derived basic protein fraction and a basic peptide fraction are provided for use as an effective component for agents for improving lipid metabolism and reducing high blood pressure which can be administered orally, are effective with a relatively small dosage, have good taste and are stable during storage.

A highly safe bone resorption suppressing agents, which can be used as medicines or admixed to food products or feeds, is produced. Milk-derived basic cystatins and/or milk-derived basic cystatin decomposition products prepared from milk are made into bone resorption suppressing agents, or admixed to drinks, food products and feeds.

A milk basic protein fraction is heated if necessary, the resulting precipitate is removed to recover the resulting supernatant, alcohol is added to this supernatant, a fraction containing a high concentration of milk basic cystatin is recovered by removing the resulting precipitate and brought into contact with a carrier containing a sulfuric acid group, such as heparin, to recover a fraction which is not adsorbed onto the carrier, and this fraction is then treated with an ultrafiltration membrane having an cut-off molecular weight of 10-50 kDa to recover the permeate to produce a fraction containing a high concentration of milk basic cystatin. Alternatively, a milk-derived basic protein composition is brought into contact with an anion exchange resin to recover a fraction which is not adsorbed onto the resin, after which this fraction is brought into contact with a cation exchange resin to recover the target fraction by eluting a fraction which is adsorbed onto the resin with an eluent to produce a fraction containing a high concentration of milk basic cystatin. Further, a decomposition product of the fraction containing a high concentration of milk basic cystatin is produced by decomposing this fraction containing a high concentration of milk basic cystatin with protease.

Method of producing fractions containing a high concentration of milk basic cystatin and decomposition products thereof

A periodontal disease preventive and ameliorative agent with milk-derived basic protein as its effective ingredient, which protein is obtained by contacting a milk or milk-derived ingredient with cation-exchange resin, and then eluting a fraction adsorbed by the resin using an elution solution and which protein has an isoelectric point within a range of 7.5&tilde;11, and food/drink and a medicament such as toothpaste and gargling agents, which contain this periodontal disease preventive and ameliorative agent.

The present invention is to provide a novel medicine, drink, food or feed having an action of strengthening bone. More specifically, the present invention is to provide a medicine, drink, food or feed combined with collagen, fraction containing collagen or degradation product thereof. Fraction containing collagen was prepared by mincing skin corium layer into pieces, defatting it and lyophylization or by pulverizing bone, decalcifying it and lyophylization. Oral administration of these can stimulate proliferation of osteoblast, inhibit bone resorption and strengthen bone. They can be useful for improvement of osteoporosis, bone fracture, bone pain, etc.

Medicine drink food and feed having an action of strengthening bone

The present invention provides a basic protein and a basic peptide composition which can be administered orally and has the ability to stimulate osteoblast proliferation, strengthen bone and inhibit bone resorption. Said basic protein has the following properties: (1) a molecular weight distribution of is 2,000-24,000; (2) an isoelectric point distribution of is 7.5-11; and (3) it is comprised of more than 10% of basic amino acids. Said composition can be obtained by the following steps: After milk, or raw material derived from milk, is loaded onto a cation exchange resin column, a fraction is obtained by eluting with an eluent of 0.1-1.0M salt concentration, precipitate removed by (i) heating to over 75.degree. C., (ii) by adding alcohol so that the final concentration thereof is 10-50% or (iii) by adding salts so that the final concentration thereof will be more than 0.2M; and the supernatant thereof containing the basic protein fraction recovered.

Basic protein composition, basic peptide composition and application

A bone reinforcing agent comprising a basic protein fraction or a basic peptide fraction derived from milk as an effective component is described. The basic protein fraction is obtained by passing milk or a raw material derived from milk over a cation exchange resin and eluting the adhered fraction. The basic peptide fraction is obtained by hydrolyzing the basic protein fraction with a protease. The basic protein fraction and basic peptide fraction of the present invention promote the growth of osteoclasts and suppress the resorption of osteoclasts, and thereby strengthening bone when administered orally. The invention is useful for treating or preventing bone diseases such as osteoporosis.

Bone reinforcing agent and foods and drinks product containing the same

The present invention will provide an agent promoting bone formation and inhibiting bone resorption comprising kininogen and degradation product of kininogen as an effective ingredient. Fragment 1.multidot.2 is a preferable degradation product of kininogen. Further, the present invention will provide a drink, food, medicine and feed combined with kininogen and degradation product of kininogen.
