The invention features nucleic acid constructs and related methods useful for the high level expression of a heterologous protein in a cell, e.g., an insect cell. The constructs include an enhancer-like sequence located upstream from the polyhedrin gene locus in the genome of baculovirus, which sequence enhances the activity of an operably linked promoter.

The invention provides a method for producing a micro-carrier, which includes patterning pluralities of bar code on a mask; exposing the bar code to a substrate coated with photoresist; etching and removing residual photoresist and electroforming to a nickel plate; placing a bead coated with biotin or poly-L-lysine between two-nickel plates, and compressing the bar code on the surface of the bead to form a microcake-like particle with bar code; and combining the particle with the corresponding bio-molecule thereof to produce a micro-carrier with a label. The invention also provides a test method for identifying a bio-molecule, which includes mixing several micro-carriers with the labeled unknown bio-molecules; and identifying the bar code on the micro-carrier via image recognition system, wherein the numbers and types of the known micro-carrier can be flexibly adjusted.

The invention relates to a baculovirus including a disruption in its endogenous p35 gene and its use in establishing latent baculovirus infections. In some embodiments, the baculovirus can also include a sequence encoding a non-baculovirus RNA and a baculovirus early gene promoter which drives expression of the non-baculovirus RNA.

Latent baculovirus expression system

Green fluorescent protein (GFP) gene was transplanted into baculovirus. The baculovirus was used to infect pests of vegetable crops. Three to five days after infection, the infected insects were found to emit strong fluorescence when exposed to a portable UV light. Therefore the spreading of baculovirus infection and disease progression can be effectively detected in the field. In addition, the GFP reduced UV damage to the baculovirus. As a result, GFP can increase the retention time of the virus as a pesticide in the field.

Use of green fluorescent protein to trace the infection of baculovirus

Disclosed and claimed is the promoter, full-length or truncated, of the persistence-associated gene 1 (pag1) of Hz-1 virus, vectors containing the promoter, including with exogenous coding DNA, methods employing the vectors, compositions employing the vectors and products therefrom, and probes and primers for the promoter or functional fragment thereof. The promoter of pag1 gene is strong and if not comparable, it is better than the promoter activity demonstrated in the polyhedrin gene in insect cells. In addition, the pag1 promoter can be expressed more prominently as a short promoter. The pag1 promoter can also be expressed in transient transfected and permanently transfected cells. Further, the pag1 promoter can also be expressed when it is inserted into other viruses, such as Autographa californica nuclear polyhedrosis virus. The pag1 promoter has also been shown to express foreign genes strongly, such as in lacZ and luciferase. pag1 is driven by a constitutively expressed early type promoter, thus facilitating expression much earlier than the polyhedrin gene promoter.
