Reducing or non-reducing polynucleotide polymer for drug delivery and method for preparing same

A system, method and device for treating tumor cells utilizing a resorbable therapy seed made up of microspheres containing a beta- or alpha-particle-emitting radiation source and a resorbable polymer matrix. These seeds are implanted within the tumor and then rapidly dissolved so as to release the microspheres from the polymer matrix. These microspheres then spread within a preselected target area and provide radiation therapy in a predetermined amount and at a preselected rate according the specific needs and necessities of the users. The configuration of the microspheres, the types of radiation provided and the location and use of these microspheres provides desired localized treatment to target cells while preferentially avoiding or minimizing undesired damage to surrounding tissue. The present invention provides a method for making the seeds, as well as a method for utilizing the seeds as a part of the treatment method.

Brachytherapy seed with fast dissolving matrix for optimal delivery of radionuclides to cancer tissue

A drug delivery vehicle for targeted delivery of a drug mimics viral properties of size, capsid-like protein capsule, cell-specific entry, toxin release, destruction of infected cells, and migration to neighboring cells. This vehicle, termed a virogel, contains a hydrophobic polymeric core, a hydrophilic inner shell, a hydrophilic outer shell, and a ligand. An illustrative drug-loaded virogel includes poly(L-histidine-co-phenylalanine) as the core, doxorubicin loaded in the core, polyethylene glycol as the inner shell, bovine serum albumin as the outer shell, and folic acid as the ligand.

The present invention relates to a block copolymer formed by coupling the following components with each other, as well as a hydrogel composition comprising the block copolymer and a hydrogel formed from the composition: (a) a copolymer of a polyethylene glycol (PEG)-based compound with a biodegradable polymer; and (b) a sulfonamide-based oligomer. The inventive block copolymer shows the sol-gel transition behavior sensitive to changes in not only temperature but also pH. Thus, the inventive block copolymer overcomes the shortcomings of temperature-sensitive copolymers, form a more strong and stable hydrogel, and is stable in vivo. Accordingly, the inventive block copolymer can be used in various applications in the medical and drug delivery fields.

PH-sensitive polymeric micelles for drug delivery

Drug delivery compositions for specific delivery of a drug to a tumor are described. These compositions include a core for sequestering the drug and a shell to which a ligand is attached for delivery of a drug to target cells. Since normal cells may also be targeted by the ligand, the compositions embed the ligand in the shell until the localized conditions surrounding the tumor cause the ligand to be displayed on the surface of the shell. One composition exhibits shrinkage of the shell at tumor pH, whereas another composition exhibits extension of the ligand at tumor pH. Still another composition causes the ligand to be exhibited at an elevated temperature.

A biosensor ( ) has a hydrogel ( ) in a rigid and referably biocompatible enclosure ( ). The hydrogel ( ) includes an immobilized glucose-binding molecule such as concanavalin A (Con A) and an immobilized hexose saccharide such as a-D-mannopyranoside. The immobilized hexose saccharide competitively binds with free glucose to the glucose-binding molecules, thus changing the number of crosslinks in the hydrogel ( ), which changes hydrogel swelling tendency and the pressure of the hydrogel in its confined space in proportion to the concentration of free glucose. By measuring the change in hydrogel pressure with a pressure transducer ( ), the biosensor ( ) is able to accurately measure the concentration of the tree glucose molecule without the problem of oxygen limitations and interference encountered by prior art biosensors. A battery ( ) powered telemeter ( ) operably engaged to the pressure transducer ( ) sends a radio data signal to a receiver ( ) containing an alarm system operably attached to a computer ( ). Furthermore, an alarm system utilizes such a sensor to automatically notify a person that the blood glucose level is outside a predetermined parameter, and/or to automatically inject an agent such as glucose or glycogen which will raise blood glucose levels.

Glucose biosensor

A biosensor (,) has a pH-sensitive polymeric hydrogel (,) in a rigid and preferably biocompatible enclosure (,). The hydrogel (,) includes an immobilized oxidoreductase enzyme such as glucose oxidase. The oxidoreductase enzyme catalyzes a chemical reaction consuming an organic molecule and producing a byproduct. The hydrogel (,) changes its osmotic pressure in proportion to the concentration of a byproduct. By measuring the change in osmotic pressure with a pressure transducer (,), the biosensor (,) is able to accurately measure the concentration of the organic molecule without the problem of interference encountered by prior art biosensors. A battery (,) powered telemeter (,) operably engaged to the pressure transducer (,) sends a radio data signal to a receiver (,) operably attached to a computer (,).

Biosensor

Terpolymers which are sensitive to pH and temperature are useful carriers for conducting bioactive agents through the gastric juices of the stomach in protected form. Such terpolymers swell at the higher physiologic pH of the intestinal tract causing release of the bioactive agents into the intestine. The terpolymers are linear and are made up of 35 to 99 wt % of a temperature sensitive component, which imparts to the terpolymer LCST (lower critical solution temperature) properties below body temperatures, 1 to 30 wt % of a pH sensitive component having a pK.sub.a in the range of from 2 to 8 which functions through ionization or deionization of carboxylic acid groups to prevent the bioactive agent from being lost at low pH but allows bioactive agent release at physiological pH of about 7.4 and a hydrophobic component which stabilizes the LCST below body temperatures and compensates for bioactive agent effects on the terpolymers. Such terpolymers provide for safe bioactive agent loading, a simple procedure for dosage form fabrication and the terpolymer functions as a protective carrier in the acidic environment of the stomach and also protects the bioactive agents from digestive enzymes until the bioactive agent is released in the intestinal tract.
