A method for detecting a substance, comprising developing a developing liquid through a test region up to a reference region, and a detection device for use therein, wherein the reference region comprises a metal compound other than an alkali metal salt, and the developing liquid that reaches the reference region contains a label that can be accumulated in the reference region. The label is preferably a colored particle, and is preferably bound to an antibody or an antigen.

Reagents for detecting antigen contained in body fluid or urine according to the immunological agglutination or agglutination inhibition reaction and comprising an antibody adsorbed on fine carrier particle to be sensitized as an effective constituent, and is characterized by acylating said antibody. Owing to such acylation, the reagent can correctly detect antigen without any influence of substance contained in sample which has inevitably caused nonspecific agglutination reaction according to the prior art.

Latex reagents for quantitative assay of a variety of steroid hormones or metabolites thereof which are contained in body fluid or excreted fluid of human beings comprise latex particles which are immunologically sensitized with a conjugate selected from a variety of steroid-serum albumin conjugates in which the steroid to be detected is bonded to serum albumin in a very small ratio ranging from 0.5 to 7 molecules per 1 molecule of the serum albumin used. Sensitization of latex particles with such a conjugate is performed using a limited amount dependent on the bonding ratio of steroid molecules per molecule of serum albumin in the conjugate and the particle size of the latex used. The latex reagents show very high sensitivity amounting to 0.2-0.04 nmole steroid equivalent/ml.
