Given developing resistance of tumor cells to current chemotherapeutic and targeted therapeutic agents, novel cancer therapies with enhanced potency and specificity are substantially required. Applicant has provided herein extracellular nanoparticle vesicles that redirect immune effector cells towards cancer cells for killing. Relative to conventional immunotherapeutic antibodies with defined orientation and geometry for their distinct antigen-binding arms, antibodies displayed on spherical exosomes can promote formation of immunological synapses as well as enhanced efficacy to activate immune cells.

Synthetic extracellular vesicles for novel therapies

This disclosure provides a method of synthesis of 4'-thioribose NAD+ and analogues thereof, using an efficient chemoenzymatic approach. Also provided are methods of inhibiting the CD38 enzyme and compounds including 4&#x2032;-thioribose NAD+ and compounds related thereto.

The connecting peptide may comprise an extender peptide. The extender peptide may have an alpha helical secondary structure. The connecting peptide may comprise a linker peptide. Coiled coil immunoglobulin fusion proteins and compositions thereof

Novel antibody-drug conjugates (ADCs) and methods of using such ADCs to treat proliferative disorders are described herein. The ADCs may comprise a PARP protein automodified with a plurality of poly-ADP-ribose polymers functionalized with novel NADanalogues. The automodified PARP with functionalized poly-ADP-ribose polymers provide a novel type of drug carrier, which allows facile conjugation of monoclonal antibodies and cytotoxic drug in high ratios.
