Engineered microorganisms for production of 2'fucosyllactose and l-fucose

The present invention relates to a method for producing marine algae-derived agarotriose, and a use thereof as a prebiotic. More specifically, the present invention investigates the characteristics of agarotriose as a prebiotic which is selectively metabolized by probiotic microorganisms, thereby enabling agarotriose to be used as an anti-cancer or anti-inflammatory agent in the fields of food and pharmaceuticals, and enabling agarotriose to be obtained at high yield through efficient purification with minimal loss after enzymatic hydrolysis of a red algae-derived polysaccharide without pre-treatment.

Method for producing marine algae-derived agarotriose, and use thereof as prebiotic

Disclosed herein are genetically modified microorganisms and related methods for enhanced utilization of oligosaccharides and improved productivity of compounds derived from the metabolism of the oligosaccharides. The microorganisms described herein have altered activities of plasma membrane ATPase protein (PMA1) and/or one or more extracellular glucose sensors, namely, sucrose non-fermenting protein (SNF3), restores glucose transport protein (RGT2), and G protein-coupled receptor 1 protein (GPR1). These genetic modifications provide the microorganisms an increased ability to utilize an oligosaccharide to produce a compound of interest, particularly, tagatose, 2'-fucosyllactose, and psicose. Methods of culturing the microorganisms in the presence of such oligosaccharides to produce the products of interest are also provided.

Engineered microorganisms for enhanced use of oligosaccharides

The present invention relates to a preparation method for various novel fucosyl oligosaccharides and a use thereof. More specifically, the present invention allows a preparation of various novel fucosyl oligosaccharides through an enzymatic reaction with &#x3b1;-1,2-fucosyltransferase using a GDP-L-fucose donor and various glucose acceptors and an establishment of probiotic characteristics thereof, and thus has an effect of providing uses as materials for medicines, food, cosmetic products, and the like.

Preparation method for various novel fucosyl oligosaccharides and use thereof

The present invention relates to methods for production of 2'Fucosyllactose by a microbial system comprising a &#x3b1;-1,2 fucosyltransferase (FucT2) polynucleotide and a Guanosine 5&#x2032;-diphospho-&#x3b2;-L-fucose (GDP-L-fucose) synthesis pathway using lactose as a substrate. Furthermore, the present invention relates to compositions comprising the microbial system.

Biosynthesis of oligosaccharides

The present disclosure provides recombinant microorganisms capable of sugar co-utilization, the recombinant microorganism comprising a genetically altered microorganism (e.g.) having a lack of, or reduced expression of, or expression of truncated or mutated forms of at least one polypeptide selected from Hxk1, Hxk2, and Glk1. Reduction of the expression or biological activity of Hxk1, Hxk2 and Glk1 leads to reduction in the glucose consumption pathway, allowing the microorganism to co-utilize multiple sugars (e.g. glucose, xylose, galactose) at an improved rate.

Recombinant microorganisms with mixed sugar utilization

The present disclosure provides genetically engineered microorganisms for tagatose production comprising one or more heterologous polynucleotides encoding polypeptides selected from cellodextrin transporter (Cdt-1), intracellular &#x3b2;-glucosidase (Gh1-1), xylose reductase (XR), galactitol 2-dehydrogenase (Gdh) and an AraA polypeptide, wherein any biological activity of endogenous Gal1 is attenuated or eliminated. Also provided are genetically engineered microorganism for psicose production comprising one or more heterologous polynucleotides encoding polypeptides selected from alpha-glucoside permease (Agt1) and psicose epimerase (Dpe), wherein any biological activity of endogenous sucrose invertase (Suc2), hexose kinase 1 (Hxk1), hexose kinase 2 (Hxk2), or combinations thereof are attenuated or eliminated. Methods of converting lactose to tagatose and sucrose to psicose is also provided.

Recombinant microorganisms for conversion of oligosaccharides into functional sweeteners

The present disclosure relates to host cells containing a recombinant polynucleotide encoding a polypeptide where the polypeptide transports cellodextrin into the cell. The present disclosure further relates to methods of increasing transport of cellodextrin into a cell, methods of increasing growth of a cell on a medium containing cellodextrin, methods of co-fermenting cellulose-derived and hemicellulose-derived sugars, and methods of making hydrocarbons or hydrocarbon derivatives by providing a host cell containing a recombinant polynucleotide encoding a polypeptide where the polypeptide transports cellodextrin into the cell. The present disclosure relates to host cells containing a recombinant polynucleotide encoding a polypeptide where the polypeptide transports a pentose into the cell, methods of increasing transport of a pentose into a cell, methods of increasing growth of a cell on a medium containing pentose sugars, and methods of making hydrocarbons or hydrocarbon derivatives by providing a host cell containing a recombinant polynucleotide encoding a polypeptide where the polypeptide transports a pentose into the cell.

Methods and compositions for improving sugar transport, mixed sugar fermentation, and production of biofuels

The present disclosure provides compositions and methods related to the fermentation of xylose. Host cells having recombinant polynucleotides encoding one or more of aldose reductases, xylitol dehydrogenases, and xylulokinase are provided herein. Host cells having reduced expression of PHO13 are also provided herein. Also provided herein are uses of host cells provided herein, and methods relating to the use of xylose-containing materials.
