A method and device for displacing fluid from a reagent cartridge () into a microfluidic device () and for loading the fluid into the reagent cartridge (). The reagent cartridge () may include a cartridge body and a pipette array with pipette tips () to engage inlets of a microfluidics or other cartridge, wherein the pipette tips () correspond in position to the plurality of inlets () of the microfluidic device (). Fluid may be loaded into or displaced from the microfluidic device () by a system of plungers (). The reagent cartridge may alternatively include blisters () having fluid reservoirs and dispensing tips (), each dispensing tip () including a pathway () that is fluidly coupled to a blister (). The fluid may be displaced from or loaded into the blister () via the dispensing tip (). A deformable seal () may be overlaid on the blisters () to seal the volumes of fluid within the blisters (), and may be deformed to displace the fluid.

Reagent cartridges for in-vitro devices

An integrated reagent cartridge can be configured to hold and deliver reagents during sequencing. The integrated reagent cartridge can include a large-volume reagent region, a small-volume reagent region, a top cover, a bottom shell assembly, and a manifold assembly.

A microfluidic device includes a substrate, a sensor, and one or more lamination films. The top surface of the substrate can include first recessed grooves forming first open channels and the bottom surface of the plastic substrate can include a first recessed cavity and second recessed groves forming second open channels. A first lamination film can be adhered with the top surface of the plastic substrate to form first closed channels. A second lamination film can be adhered to the bottom surface of the plastic substrate to form second closed channels. The sensor can be on the bottom surface of the substrate such that it overlies the first recessed cavity to form a flow cell with the sensor top surface inward facing. A first closed channel can be fluidically connected with a second closed channel and a first or second closed channel can be fluidically connected with the flow cell.
