Porcine pestivirus designated herein as atypical porcine pestivirus ('APPV') (Genbank accession no. KR011347.1). Immunogenic compositions to induce an immune response against porcine pestivirus infection in a pig are described, which APPV antigenic agents (e.g., isolated whole virus, derivatives thereof, functional fragments thereof, and combinations of the foregoing). Methods of vaccinating against porcine pestivirus infection using the immunogenic compositions are also described. The methods can be also applied for clinical research and/or study, including diagnostic methods for detecting pestivirus infection using monoclonal antibodies specifically binding to APPV epitopes.

Porcine pestvirus, vaccines, and assays

Disclosed herein are compositions and methods useful for immunizing a subject against disease caused by influenza A. Disclosed methods comprise administering to the subject an immunoprotective dose of an immunogenic composition. In certain aspects, the immunogenic composition is a vaccine comprised of a recombinant chimeric hemagglutinin polypeptide. In certain aspects, the subject is a mammal. In further aspects, the mammal is a pig. In still further aspects, the mammal is a human.

Compositions and methods for vaccination against influenza A virus

The present invention relates to modified, live Porcine Reproductive and Respiratory Syndrome viruses. Viruses were genetically analyzed and selected based on phylogenetic grouping for modification by repeated passage in tissue culture. The modified, live viruses were assessed for the ability to provide protective immunity to heterologous viruses. The modified, live viruses are useful in vaccines, particularly in vaccines which can treat infection of swine by multiple heterologous viruses.

Porcine reproductive and respiratory syndrome vaccine virus

The invention provides the discovery and characterization of a novel arterivirus protein (nsp2TF), whose expression is dependent on &#x2212;2 ribosomal frameshifting at a site located in the nsp2 coding region. The coding region for the unique TF domain of nsp2TF overlaps the part of ORF1a that encodes the transmembrane region of nsp2 in arteriviruses, including PRRSV, LDV and SHFV. Mutations affecting the expression of nsp2TF impair PRRSV replication and result in a smaller plaque phenotype. Provided herein are arteriviruses that display reduced translation of nsp2TF and/or altered translation of one or more downstream products, arteriviruses in which nsp2TF function is reduced and/or absent, and vaccines comprising said arteriviruses. Also provided herein are diagnostic methods, methods for identifying compounds that inhibit &#x2212;2 frameshifting, and gene expression tools for eukaryotic systems utilizing &#x2212;2 frameshifting.

Arterivirus protein and expression mechanisms

Porcine reproductive and respiratory syndrome virus (PRRSV) is a major problem in the pork industry worldwide. The inclusion of markers in vaccines will allow for diagnostic differentiation of vaccinated animals from those naturally infected with wild-type virus. Using a cDNA infectious clone of North American Type 1 PRRSV, a recombinant green fluorescent protein (GFP) tagged PRRSV has been made, containing deletion of an immunogenic epitope, ES4, in the nsp2 region. GFP and ES4 epitope-based ELISAs compliment the marker identification.
