The efficacy of a dosing regimen can be altered, and preferably improved, by fractionating the doses. Here fractionating the doses can be regarded as taking a single conventional dose and splitting that dose into two or more dose-fractions delivered at relatively short time intervals, where the dose-fractions add up to the same total dose as the conventional dose. In cases where dose fractionation increases the therapeutic effect, this can result in a greater therapeutic effect from a smaller total dose when the doses are fractionated. This effect has been demonstrated in an ophthalmic application, but is expected to be more generally applicable.

Fractionated topical ocular drug delivery methods and devices for use in the same

Multi-dose ocular fluid delivery devices are provided. Aspects of the fluid delivery devices include a fluid package and an actuator. The fluid package includes a reservoir of an ophthalmic formulation, an aperture and a valve member for sealing the aperture when fluid is not being ejected therethrough. The actuator operates the valve member so as to at least reduce, if not prevent, ingress of outside materials or contaminants into the reservoir. Also provided are methods of using the devices in fluid delivery applications, as well as a kit that includes components of the devices.

Multi-dose ocular fluid delivery system

Micro-droplet delivery devices and methods are described where the device may comprise a piezoelectric actuator having a piezoelectric chip that is operatively coupled to a drug package under a preloading force. The actuator is configured to generate an acoustic pressure within the drug package to dispense droplets of an agent from an aperture, e.g., to the corneal surface of the eye. The piezoelectric actuator can be coupled or decoupled from the drug package.

Multi-dose preservative-free ocular fluid delivery devices are provided. The fluid delivery device includes a fluid dispensing system and a fluid package for storing a liquid therein and supplying said liquid to the dispensing system. The dispensing system comprises an elongated chamber which includes a check valve which defines a frontal closure to the chamber. The valve is normally closed and hermetically seals the chamber. The dispenser includes a vibration motor that induces oscillations to the chamber and to the fluid within. The oscillations of the chamber impart momentum to the fluid stored in the chamber which in turn imparts force that cyclically opens the valve to dispense streams or liquid droplets. Fluid is dispensed only when the motor oscillates while otherwise the valve is hermetically closed.

Hydrodynamically actuated preservative free dispensing system

Methods of administering a liquid formulation of an ophthalmic agent to a topical ocular location of an eye are provided. Aspects of the methods include delivering to the topical ocular location a dose of the liquid formulation that can be wholly accommodated by the tear film of the eye. Devices and kits for practicing the methods are also provided. The methods, compositions and kits find use in a variety of applications, including therapeutic, diagnostic and cosmetic applications. Further aspects include wireless communication between a handheld dispenser of the ophthalmic agent and external devices(s) to provide various functions.

Topical ocular delivery of fluids with controlled mass dosing and wireless communication

Topical ocular delivery devices are provided. Aspects of the devices include a transducer configured to oscillate at an amplitude and frequency that provides for an emitted stream velocity which results in minimal discomfort during topical ocular delivery. Also provided are methods of using the devices, e.g., in topical ocular delivery applications.

Aspects of the invention include a device for fluid delivery to the ocular surface having a self-alignment system. The fluid delivery device includes a fluid package having a reservoir and one or more apertures, an actuator component configured to eject fluid from the reservoir through the one or more apertures and an image-based self-alignment system configured to align fluid ejected through the one or more apertures with a target location on the eye of a user. A device for delivery of a precise amount of pharmaceutical fluid to the eye is provided. The device includes an ampoule for storing a liquid and an aperture through which the liquid is discharged. The device further includes a vibrating membrane that includes a needle which protrudes from the membrane and extends to the aperture to form a needle valve. The needle valve provides hermetic closure of the aperture to enable preservative-free storage of the pharmaceutical fluid in the device. The system further includes an electromagnetic transducer to shift the membrane back and forth in order to discharge liquid through the nozzle. Finally, the system further includes a vent to permit air to enter the ampoule as fluid is ejected from the ampoule. Preferably this vent includes a filter to prevent ingress of microbes or other contaminants to the ampoule.

Fluid delivery devices and methods are described, where the device may comprise a piezoelectric actuator operatively coupled to an ampoule with fluid, under a preloading force. The ampoule may comprise a thin-walled thermoplastic package which includes one or more apertures positioned on the wall of the ampoule. The piezoelectric actuator may be configured to clamp at least partially around the circumference of the ampoule and apply oscillations to its wall. The oscillations, typically in ultrasonic frequency, produce cycles of acoustic pressure in the fluid, leading to ejection of the fluid droplets or streams from the apertures.
