An electrode for electroporation comprising a plurality of electrode needles, wherein first polarity electrode needles, an electrode needle holding portion, and a syringe holding portion are provided; two or more first polarity electrode needles project from a bottom surface of a lower structural body of an outer frame support of the electrode needle holding portion toward an electroporation target side; the bottom surface of the lower structural body of the outer frame support is provided with a hole for syringe needle insertion and removal communicating with a syringe holding portion side; the syringe holding portion is provided on a side opposite to the electroporation target side of the electrode needle holding portion; and the syringe holding portion has a path for syringe needle insertion and removal, at least a portion of the path for syringe needle insertion and removal being provided with an electro-conductive portion for a second polarity.

Electrodes for electroporation

An object is to develop a technology enabling utilization, by only an extremely simple technique, of a technology which is widely applicable to mammals without requiring the utilization of an ES cell, and which involves modifying a certain gene by targeting a certain sequence on a genome (genome editing technology based on ZFN or the like). Provided is a technology for efficiently modifying an arbitrary target gene of a mammal, by immersing a pronuclear stage mammalian zygote with an intact zona pellucida into a solution containing a pair of molecules of mRNA having a certain sequence, and performing electroporation treatment through application of multiple square-wave pulses in three steps with the total electric energy of a first electric pulse adjusted within a predetermined range.

Mammalian gene modification method using electroporation

An electric pulse generator for electroporator includes a poring pulse generator, and a transfer pulse generator. The poring generator includes an n-stage Cockroft-Walton circuit, and a first circuit branched from a branching point of wiring on output side of the Cockroft-Walton, the first circuit includes a switching switch that is turned off in high-voltage mode and turned on in low-voltage mode, a voltage value for switching between the modes is in 200 to 1400 V, the first circuit includes a second circuit in which mseries of mseries-connected capacitors are connected in parallel, the switch and second circuit are connected in series, and first wiring on output side of the second circuit is merged with second wiring on output side of the Cockroft-Walton on output side of the branching point, and a withstand voltage of the Cockroft-Walton is a value in 1500 to 5000 V in the high-voltage mode.

A method of transferring an exogenous gene into a eukaryotic algal cell, the method including performing electroporation using multiple square-wave pulses to the solution containing a cell of a green alga with cell-wall, and a nucleic acid molecule by the following three steps: applying a square-wave electric pulse (first electric pulse) with a high voltage for a short period of time under the condition that its total electric energy falls within a predetermined range; then applying a square-wave electric pulse (second electric pulse) with a low voltage for a long period of time two or more times; and then applying a square-wave electric pulse (third electric pulse) that is opposite in polarity to the second electric pulse, with a low voltage for a long period of time, two or more times.

Method of transferring gene into algal cell involving utilizing multiple square-wave pulses in three steps

A method for in vivo electrotherapy, or electroporation-mediated therapy, using a needle array apparatus is provided. Treatment of tumors with a combination of electroporation using the apparatus of the invention, and a chemotherapeutic agent, caused regression of tumors in vivo.

A method for producing a genetically modified plant by introducing a polynucleotide to an intact plant or plant cell(s) via electroporation, in the absence of cell wall-degrading enzymes. Genetically engineered plants produced by the method of the invention are also provided. The invention also provides a method for producing a polypeptide in an intact plant cell, including plant tissue or a whole plant by introducing a biologically active polypeptide directly into the plant.

Electroporation-mediated molecular transfer in intact plants

An electrode apparatus for the application of electroporation to a portion of the body of a patient, comprises a support member, a pair of electrodes adjustably mounted on the support member for movement toward and away from one another for positioning on opposite sides of a body portion to be electroporated, a sensing element for sensing a distance between the electrodes and generating a distance signal proportionate to the distance between said electrodes, and means responsive to said distance signal for applying pulses of high amplitude electric signal to the electrodes proportionate to the distance between said electrodes.
