A system for playback of messages. Context appropriate messages for an environment may be played back. Messages may be user behavior interactive and subject to user behavior initiated message playback conditions. User generated environment events may be automatically analyzed and user behavior interactive messages may be automatically coordinated. An automated themed message playback apparatus may have a self-contained housing within which a stored themed message, an in situ user generated environment event sensor, and an automated themed message playback device are housed. User generated environment events may be automatically sensed in situ.

Varied apparatus for message playback

Provided are methods for labeling transfer RNA comprising replacing the uracil component of a dihydrouridine of said transfer RNA with a fluorophore. The disclosed methods may comprise fluorescent labeling of natural tRNAs (i.e., tRNAs that have been synthesized in a cell, for example, in a bacterium, a yeast cell, or a vertebrate cell) at dihydrouridine (D) positions, or fluorescent labeling of synthetic tRNAs. In another aspect, the present invention provides methods for assessing protein synthesis in a translation system comprise providing a tRNA having a fluorophore substitution for the uracil component of a dihydrouridine in a D loop of the tRNA; introducing the labeled tRNA into the translation system; irradiating the translation system with electromagnetic radiation, thereby generating a fluorescence signal from the fluorophore; detecting the fluorescence signal; and, correlating the fluorescence signal to one or more characteristics of the protein synthesis in the translation system. The disclosed methods are useful in single molecule as well as in ensemble settings.

Catheter systems for measuring at least one electrical property, e.g., impedance, of cardiac tissue of a living being are disclosed. The system includes a catheter having a tip with a sensing electrode, a guard electrode and an electrical shield. The sensing electrode is arranged to engage the cardiac tissue and is coupled to circuitry for measuring the at least one electrical property of the cardiac tissue, shielding the sensing electrode from bulk blood adjacent the cardiac tissue. The measurement can gated to the cardiac cycle. Additional embodiments include multi-electrode sensor catheter tips for high density mapping. Moreover, such tips may be dynamically configurable, i.e., their electrodes can be variably assigned as sensor electrodes or guard electrodes by associated circuitry. Such multi-electrode configuration and reconfiguration can be gated to the cardiac cycle.
