Provided are a fluorescent dye and a preparation process and use thereof. The fluorescent dye is able to emit a long wavelength, is sensitive and specific to viscosity, and is used for a wide range of purposes, such as viscosity testing, fluorescent labeling, quantification or detection of proteins, enzymes or nucleic acids. In addition, it can be used as a fluorescent-activated probe.

Provided are a fluorescent probe and a preparation process and the use thereof. The fluorescent probe is sensitive and specific to viscosity, and can be used for specific fluorescent labeling of proteins, and can also be used for quantification, detection or kinetic studies of proteins, and the imaging of cells, tissues and living bodies.

Fluorescent probe and preparation method and use thereof

Provided is an optically controlled gene expression system of prokaryotic bacterium, comprising: a) a photosensitive recombinant transcription factor encoding gene, the photosensitive recombinant transcription factor is one fusion protein comprising a first polypeptide as the DNA bonding domain and a second polypeptide as the photosensitive domain; b) a target transcription unit comprising promoter or promoter-reaction element or reaction element-promoter containing at least one reaction element recognized/bound by the first polypeptide and the nucleic acid sequence to be transcribed. Also provided is a prokaryotic expression vector comprising said optically controlled gene expression system, and a method for regulating gene expression in a prokaryotic host cell by using the optically controlled gene expression system. Also provided is a reagent kit containing different components of the optically controlled gene expression system. The optically controlled gene expression system of prokaryotic bacterium has a quick, effective and powerful induction, is safer than other inducers, is of little or no toxicity, and can control gene expression both spatially and temporally, and can regulate many life processes of prokaryotic bacterium.

Light-switchable gene expression system and the methods for controlling gene expression in prokaryotic bacterium

Provided is a light-switchable gene expression system, comprising: a) a recombinant light-switchable transcription factor-encoding gene, said recombinant light-switchable transcription factor comprising a first polypeptide as the DNA bonding domain, a second polypeptide as the photosensitive domain, and a third polypeptide as the transcription regulatory domain; b) a target transcription unit comprising at least one reaction element recognized and bound by the first polypeptide, a promoter regulated by the third polypeptide and a nucleotide sequence to be transcribed. Also provided is an eukaryotic expression vector comprising said light-switchable gene expression system, and a method for regulating gene expression in a host cell by using the light-switchable gene expression system. Also provided is a reagent kit containing different elements of the light-switchable gene expression system. The light-switchable gene expression system has a quick, effective and powerful induction with little or no toxicity. It is safer than other inducers, and can spatiotemporally control gene expression.
