The present invention relates to substrates for biological testing produced from photo-curable epoxy compositions which further include carboxyl-containing monomers such as acrylic acid, 2-carboxyethyl acrylic acid, 4-vinylbenzoic acid, or 3-Acrylamido-3-methyl-1-butanoic acid, or glycidyl methacrylate, etc. The photo-curable compositions may be used to cast films or fabricate beads, magnetic beads, or magnetic beads containing nickel barcodes. The resulting various kinds of films, beads, magnetic beads, or magnetic beads containing nickel barcodes may find use in clinical or biological applications.

Biocompatible and photocurable polymers

A micro bead having a digitally coded structure that is partially transmissive and opaque to light. The pattern of transmitted light is determined by to decode the bead. The coded bead may be structured a series of alternating light transmissive and opaque sections, with relative positions, widths and spacing resembling a 1D or 2D bar code image. Alternative method for barcode determination is based on barcode pattern image processes. Microbeads can also be settled down to the bottom of the well in a microplate, so the barcode can be decoded by image processed directly. Therefore, multiple-analyte tests can be performed in one single well. Microplate is a standard format; each plate can have 96, 384, or 1536 patient samples. The bead image is taken with a conventional microscope and optical detector from the bottom of an optically clear microplate, and barcode pattern can be decoded by image software. Therefore, the whole bead experiment can be performed in the microplate without taking the beads out.

The method for accurately counting photon numbers entering a photon multiplier tube comprising the steps of counting the number of the electrical pulses generated by photons; measuring the duration of the electrical pulses with a timing clock, wherein the timing clock can count the number of temporally overlapped photons; measuring the intensity of the electrical pulses with an intensity discriminator, wherein the intensity discriminator can count the number of intensity-overlapped photons; and summing the number of the electrical pulses, the number of temporally overlapped photon, and the number of intensity-overlapped photons, that is equal to the total number of photons.

An optical apparatus and methods for monitoring temperature enabling detection and treatment of a vulnerable plaque of a patient comprising an elongate catheter, a plurality of outer optical fibers deployably disposed within the lumen of the catheter and suitably expandable in an outwardly radial manner configured for forming a basket shape, the outer fibers having at least one optical grating along an axis of the fiber wherein the at least one optical grating reflects a certain wavelength or intensity of the light beam, the certain wavelength or intensity of the reflected light beam being correlated to known temperature, and a longitudinal middle optical fiber emitting a light energy suitable for photodynamic therapy.

Optical apparatus for detecting and treating vulnerable plaque

A biochip and apparatus is disclosed for performing biological assays in a self-contained microfluidic platform. The disposable biochip for multi-step reactions comprises a body structure with a plurality of reagent cavities and reaction wells connected via microfluidic channels; the reagent cavities with reagent sealing means for storing a plurality of reagents; the reagent sealing means being breakable and allowing a sequence of reagents to be released into microfluidic channel and reaction well; and the reaction well allowing multi-step reactions to occur by sequentially removing away the residual reagents. The analysis apparatus can rapidly, automatically, sensitively, and simultaneously detect and identify multiple analytes or multiple samples in a very small quantity.

The disclosure describes how to use luminescence detection mechanism, move microfluid, and control multiple-step biochemical reactions in closed confined microfluidic biochip platform. More particularly, a self-contained disposable biochip with patterned microchannels and compartments having storage means for storing a plurality of samples, reagents, and luminescent substrates. At least one external microactuator in the biochip system produces positive pressure and automates multiple-step reactions in microfluidic platforms for clinical chemistry, cell biology, immunoassay and nucleic acid analysis. The method comprises the steps of transferring sequentially at least one of samples, reagents, and then luminescent substrate from compartments through microchannels to reaction sites. The luminescent substrates react with probes to form a probe complex resulting into luminescence, which is detected by an optical detector.

An on-site monitor is disclosed for providing automatic measurements of chemical concentrations in a liquid or gaseous environment. The opto-electronic monitor has an extended probe, which can be inserted into the liquid or to be in contact with the ambiance, comprising a indicator chamber for continuously supplying fresh indicator material, a light source, and a detector. The light source and the detector are either embodied outside of liquid or into the extended plastic probe inside the liquid. A delivery unit, which is activated by a timer, delivers a small amount of indicator material into the liquid or a sampling cavity. The indicator material reacts with the ions in the liquid automatically. The light source has a wavelength which matches with the indicator's absorption wavelength and the detector measures the transmission (or reflection) through the indicator material or solution. The optical signal is converted into an electrical signal, calibrated with a known standard, and then displayed on an LED digital meter. The invention has the advantage of automatically and continuously measuring the chemicals on-site with self-powered, low maintenance, and simple operation.
