The invention provides methods for overcoming glucocorticoid resistance of cancers by inhibition of CASP1. Also disclosed are diagnostic methods for determining glucocorticoid resistance potential by measuring expression level or promoter methylation status of CASP1 gene and/or NLRP3 gene.

Methods for overcoming glucocorticoid resistance and for determining glucocorticoid resistance potential in cancer

Single nucleotide polymorphisms (SNPs) in the gene encoding gamma glutamyl hydrolase (GGH) associated with reduced GGH activity are disclosed. The primary SNP is a change from a cytosine to a thymine at a position corresponding to nucleotide 511 of Genbank sequence accession no. NM 003878. Methods and kits for detecting these SNPs are provided, along with primers useful in detecting these SNP and for amplifying portions of the GGH gene containing these SNPs.

Genotyping assay to predict gamma glutamyl hydrolase (GGH) activity

The invention relates to methods for detecting genetic polymorphisms in an organism, particularly to the detection of genetic polymorphisms that are due to multiple distal nucleotide polymorphisms within a gene. Methods are provided for determining the haplotype structure of a gene, or other contiguous DNA segment, having two or more nucleotide polymorphisms that are separated by kilobases of DNA. The methods involve the use of PCR amplification and DNA ligation to bring the nucleotide polymorphisms on a particular allele of the gene into close proximity to facilitate the determination of haplotype structure.

Haplotyping method for multiple distal nucleotide polymorphisms

Mutants of thiopurine S-methyltransferase (TPMT) are described. TPMTA mutant has a point mutation at cDNA position 238 (G.sup.238 .fwdarw.C), and TPMTB involves two nucleotide transitions at cDNA positions 460 (G.sup.460 .fwdarw.A) and 719 (A.sup.719 .fwdarw.G). TPMTB is the predominant mutant allele associated with human TPMT-deficiency which can cause potentially fatal toxicity when patients are treated with mercaptopurine, azathioprine, or thioguanine. The mutant alleles as well as PCR fragments, mutant proteins and antibodies therefor, together with kits and methods for assaying the TPMT genotype of individual patients are disclosed.

Identification of two novel mutant alleles of human thiopurine

An integral molded metal sinker has a streamlined body with a pair of wings and a keel fin projecting therefrom. The body has a bulbous, semi-spherical front end which tapers back smoothly to a relatively small rear end. A wire is embedded in the body and extends from the rear end to the front end thereof. Swivels are connected to opposite ends of the wire for attaching the sinker to a fishing line and to a leader. The rear swivel is near the central axis of the body, but the front swivel is substantially above the central axis of the body, thereby placing the center of gravity of the body substantially below the front swivel and tending to prevent rotation of the body. The leading edge portions of the wings are angled downwardly with respect to the rear portions to make the sinker tend to dive when it is drawn through the water. The downturned wing portions are substantially rectangular in shape to maximize their area for a given wing span and width.
