A novel agarose gel is provided for use in the high resolution electrophoretic (HRE) separation of serum proteins. The agarose gel employs a novel gel buffer which contains either hippurate, glycine, or a mixture thereof, in combination with barbital and Tris. The agarose gel of the present invention yields improved electrophoretic separation of the serum proteins in a sample whether the same gel buffer is also used as the running buffer or whether a standard barbital running buffer is used.

The electrophoretic gel comprises a matrix selected from the group consisting of polysaccharides and derivatives thereof, a buffer having a buffering capacity in an alkaline pH range, and an effective amount of a poly(oxyethylene)-poly(oxypropylene)-poly(oxyethylene) block copolymer. An improved electrophoretic technique characterized in that the above electrophoretic gel is employed therein.

An electrophoretic technique for assaying the relative distribution of lactate dehydrogenase (LD) isoenzymes characterized in that the electrophoretic and/or substrate buffer employed therein comprises a first and second moiety wherein the first moiety is selected from a group consisting of alkali metal 5,5-diethylbarbiturate, ammonium 5,5-diethylbarbiturate, and mixtures thereof, and wherein the second moiety is selected from a group consisting of bicine (N,N-bis(2-hydroxyethyl)glycine), tricine (N-tris(hydroxymethyl)methylglycine), and glycylglycine. The buffer has a pH of from about 7 to about 9 at 25.degree. C. Preferably, the buffer further comprises a solid organic water soluble acid and either 2-amino-2-methyl-1,3-propanediol (AMPD) or 2-amino-2-hydroxymethyl-1,3-propanediol (Tris) or mixtures thereof. Buffers within the scope of the instant invention, when compared to prior art buffers, increase the relative percentage of LD.sub.5 and thereby improve the accuracy and reproducibility of the electrophoretic data.

A method of clearing a polysaccharide ester membrane employing a clearing solvent comprising a first moiety selected from a group consisting of alcohols, ketones, ethers, esters, and mixtures thereof and a second moiety comprising a solvent selected from a group consisting of 2-ethoxyethanol, 2-ethoxyethyl acetate, methyl acetoacetate, ethyl acetoacetate, acetol, and mixtures thereof, the first moiety having a boiling point lower than the second moiety. The solvents employed as the second moiety of the instant invention's clearing solvent are much less toxic than solvents employed as the second moiety of prior art clearing solvents.
