The invention relates to a microfabricated device and methods of using the device for analyzing and sorting polynucleotide molecules by size.

Method and apparatus for analysis and sorting of polynucleotides based on size

The present invention provides an apparatus for analyzing the sequences of polynucleotides. The apparatus comprises (a) flow cell which has at least one microfabricated multilayer elastomeric synthesis channel; and (b) an inlet port and an outlet port. The inlet port and outlet ports are in fluid communication with the flow cell for flowing fluids into and through the flow cell.

Methods and apparatus for analyzing polynucleotide sequences

The invention provides compositions and novel polynucleotides and proteins that co-express with known marker genes for inflammatory disorders. The invention also provides expression vectors, host cells, proteins encoded by the polynucleotides and antibodies which specifically bind the proteins. The invention also provides methods for the diagnosis, prognosis, evaluation of therapies and treatment of inflammatory disorders.

Inflammation-associated polynucleotides

The invention provides a mammalian cDNA which encodes a mammalian AQP8V. It also provides for the use of the cDNA, fragments, complements, and variants thereof and of the encoded protein, portions thereof and antibodies thereto for diagnosis and treatment of pancreatic disorders, particularly type I diabetes. The invention additionally provides expression vectors and host cells for the production of the protein and a transgenic model system.

Aquaporin-8 variant

The invention provides polynucleotides and polypeptides that are co-expressed with genes known to be involved in bone remodeling and osteoporosis. The invention also provides expression vectors, host cells, and methods for making a polypeptide, for screening or purifying ligands, and for diagnosing disorders associated with bone remodeling or osteoporosis.

The invention relates to a microfabricated device and methods of using the device for analyzing and sorting individual polynucleotides, e.g., by size according to an optical signal measured within a detection region of the device. An optical signal such as fluorescence from a reporter molecule associated with the polynucleotide molecules can be used to determine polynucleotide size or to direct selected polynucleotides into one or more selected branch channels of the device.

Methods for analysis and sorting of polynucleotides

The invention provides novel kidney disease-associated genes and polypeptides encoded by those genes. The invention also provides expression vectors, host cells, and antibodies. The invention also provides methods for diagnosing, treating or preventing diseases of the kidney.
