Isolated polypeptides, nucleic acids, and methods relating to cellular internalization of materials are described herein. Generally, the isolated polypeptides include a membrane transduction domain of human tissue factor pathway inhibitor-2 (TFPI-2). In some cases, the isolated polypeptide can be a fusion peptide that includes a membrane transduction domain of human TFPI-2 and a heterologous peptide domain. The nucleic acids include nucleic acids that encode the isolated polypeptides described herein. The methods generally include providing a composition that includes a membrane transduction domain of human TFPI-2 coupled to a material, and contacting the composition with a cell under conditions effective to permit the cell to internalize the composition.

Membrane transduction peptides and methods of delivering material to a target cell

A human Kunitz-type inhibitor polypeptide with enhanced antifibrinolytic activity, methods of making, and methods of use. The novel polypeptide is structurally similar to the KD1 domain of human tissue factor pathway inhibitor-2 (TFPI-2). In another aspect, methods of treating a subject afflicted with cancer or a precancerous condition are described. Generally, the method includes administering to a subject in need of treatment an effective amount of a polypeptide. In some embodiments, the polypeptide comprises a KD1 domain of human TFPI-2. In some embodiments, the polypeptide comprises human TFPI-2, itself. In certain embodiments, the polypeptide is administered in an amount effective to induce apoptosis in tumor cells.

Human kunitz-type inhibitor with enhanced antifibrinolytic activity

The invention includes compositions, kits and methods comprising a monoclonal antibody which shares key functional properties with the polyclonal antibodies which participate in the pathogenesis of heparin induced thrombocytopenia/thrombosis (HIT/HITT) in a mammal. The monoclonal antibody of the invention preferentially binds with a PF4/heparin complex relative to the binding of the antibody with PF4 or heparin alone. The monoclonal antibody of the invention also binds specifically with PF4 in a complex with other glycosaminoglycans besides heparin, and also activates platelets. The monoclonal antibody of the invention is useful in methods for diagnosing and treating HIT/HITT in a mammal. A humanized version of the monoclonal antibody of the invention is also included, along with a process for humanizing the monoclonal antibody of the invention.
