The present disclosure relates to recombinant therapeutic cells comprising recombinant sensor proteins, such that a therapeutic molecule is expressed after the occurrence of two separate triggering events that are sensed by the sensor proteins. Nucleic acids, kits, and methods for making and using the recombinant therapeutic cells are also disclosed herein.

Conditional modulation of therapy using recombinant cells

Described herein are methods of making recombinant cells using an in silico-generated pathway map of an endogenous pathway in a cell of interest to determine, in silico, a predicted effect of incorporating a recombinant nucleic acid into the endogenous pathway; and based on the predicted effect, incorporating the recombinant nucleic acid into the endogenous pathway in the cell of interest such that activation of the endogenous pathway regulates expression of the recombinant nucleic acid.

Prediction of computational pathway circuits

Compositions and methods are presented that allow for an enhanced immune response against a GPI-anchored tumor associated antigen by modification of the protein portion of the TAA to include a transmembrane domain and a trafficking signal that directs the modified protein to the endosomal or lysosomal compartment. Method of detecting cellular reporting for therapy production

Recombinant cells and methods therefor are contemplated that allow for rapid and high titer production of recombinant viruses, and especially replication deficient Ad5 virus. In some preferred aspects, the host cell is modified to produce an inhibitor that reduces or eliminates the expression of a therapeutic protein encoded in the virus, while in other aspects, the virus includes a gene that directly or indirectly reduces or eliminates the expression of a therapeutic protein encoded in the virus. Most preferably, shRNA encoded by the host cell will reduce or suppress expression of a payload gene encoded in the recombinant virus.
