The present invention provides an intravenous liposomal formulation of a low dose polymyxin B or a pharmaceutical composition thereof. The present invention also provides methods utilizing the intravenous liposomal formulation or its pharmaceutical composition for increasing the therapeutic efficacy during treatment of a bacterial infection in a subject, for decreasing exposure of renal tissue to the polymyxin B during a course of polymyxin B treatment and for lowering exposure to polymyxin B in renal tissues in a subject receiving polymyxin B treatment.

Liposomal formulations of polymyxin B and uses thereof

The present invention provides novel liposomal formulations of polymyxin B and pharmaceutical compositions thereof useful for the treatment of bacterial infections. The liposomal formulation comprises a lipid component formed as vesicles each having a minimum size of at least 500 nm and polymyxin B encapsulated in the vesicles. The present invention also provides a drug delivery system that comprises a plurality of liposomes encapsulating a polymyxin B therein, where the liposomes have a minimum vesicular size of at least 500 nm. The liposomes comprising the drug delivery system are useful to increase efficacy of a treatment for a bacterial infection by increasing bioavailability and distribution of the polymyxin B within the subject.

Liposomal formulations of polymyxin and uses thereof

Provided herein are methods and computer-implemented systems for using computer simulations to predict likelihood of a cell population associated with a pathophysiological condition acquiring resistance to a therapeutic agent, to screen for therapeutic agents effective to suppress acquisition of resistance within a cell population and to treat the pathophysiological conditions associated therewith. The computer simulation comprises at least an input/out system and a mathematical model, including operably linked equations, parameter values and constant values, of growth response over a period of time of a cell population in contact with an therapeutic agent. Also provide is a method for determining a best-fit mathematical model of adaptation of a microbial population to a therapeutic agent over time and using the model to simulate microbial population behavior to a therapeutic agent.
