Provided herein are compositions and methods of using Seneca Valley Virus (SVV) or a derivative thereof and an interferon type I (IFN-I) inhibiting agent comprising an mTOR inhibitor for treating a cancer in a subject. The disclosed methods particularly rely upon the expression level of an ANTXR1 and the expression level of IFN-I in the cancer from the subject. Also provided herein are methods for predicting the efficacy of an SVV treatment comprising IFN-I inhibiting agent.

Second generation Seneca Valley virus oncolytic therapy: compositions and methods thereof

The present invention relates to methods for producing RNA virus cDNA, methods for producing viable, RNA virus and viable, RNA virus produced by those methods. The invention also related to a novel RNA virus cDNA, recombinant DNA molecules containing that cDNA and hosts transformed with those recombinant cDNA molecules. This invention further related to novel methods for screening for variants of a strain 3 poliovirus. This invention also related to methods for increasing the attenuation of a strain poliovirus. This invention provides a vaccine useful for immunizing a subject, for example a human, against infectious poliovirus, wherein the vaccine comprises an effective amount of an RNA virus, produced by transforming a suitable host cell with a recombinant nucleic acid sequence which encodes for the virus: culturing the host cell under conditions which permit the production of virus: and isolating the virus so produced, effective to immunize the subject, and a suitable carrier. Further provided to this invention is a method of immunizing a subject such as a human against infectious poliovirus, wherein the method comprises administering the subject a suitable dose of the vaccine described hereinabove.

The subject invention provides an isolated nucleic acid molecule which comprises nucleic acid encoding a polypeptide which has the biological activity of a receptor for poliovirus and in addition, provides a purified polypeptide which has this activity. The subject invention also provides a method for inducing the production of a polypeptides which includes the use of expression vectors in a host vector system. Therapeutic compositions comprising purified polypeptides which have the biological activity of a receptor for poliovirus are also provided for as are methods of treating and preventing human poliovirus infection. Further, the subject invention provides a method of producing a transgenic animal expressing human poliovirus receptors. The subject invention provides a method of testing the efficacy of a poliovirus receptors vaccine which comprises administering the vaccine to a transgenic animal described above and determining whether the resulting transgenic animal is protected from infection by human picornavirus. Lastly, a method of testing the virulence of a picornavirus vaccine is provided.

Methods for producing HAV cDNA, products thereof, and uses thereof, are described. HAV cDNA is produced, for example, by reverse transcribing HAV RNA and subsequently inserting the HAV cDNA into bacterial plasmids by genetic-engineering techniques. Transformed bacteria are then cloned and cultured to produce replicated chimetic plasmids containing the HAV cDNA. Such HAV cDNA is useful in assaying for the presence of HAV and in the production of HAV antigen and in the production of antibodies against HAV.
