The invention provides: a cross-linkable therapeutic composition comprising a) a saccharide-siloxane copolymer; b) a crosslinking agent; c) one or more active/inactive ingredients; and d) optionally, a solvent, or solvent mixture, wherein the saccharide-siloxane copolymer has the following formula: RaR(3-a)SiO&#x2014;[(SiRRO)m-(SiR2O)n]y-SiR(3-a)Ra that is further formulaically defined and wherein the saccharide-siloxane copolymer is a reaction product of a functionalized organosiloxane polymer and at least one hydroxy-functional saccharide such that the organosiloxane component is covalently linked via a linking group to the saccharide component; films; and methods related thereto. The composition has therapeutic properties. The invention also provides articles of manufacture including topical and transdermal agent delivery patches comprising the novel film-forming composition and/or films.

Silicone film former for delivery of actives

This invention pertains to neutralized silicone elastomer dispersions and methods for making neutralized silicone elastomer dispersions. This invention also pertains to compositions comprising the neutralized silicone elastomer and an acid reactive compound. The silicone elastomer dispersion are neutralized typically by blending the silicone elastomer dispersion with a base such as sodium bicarbonate or by adding a basic neutralizing agent to one or more of the raw materials used to make the silicone elastomer dispersion and removing the basic neutralizing agent by filtration or any other suitable means.

Stabilized elastomer dispersions

A controlled-release composition for topical application to a substrate includes an oil-in-water emulsion and an active agent incorporated into the oil-in-water emulsion. The oil-in-water emulsion is substantially free of lipophilic solvent and is formed by mechanical inversion of a water-in-oil emulsion. The water-in-oil emulsion includes a silicone component, a surfactant, and water. A method of delivering the active agent to the substrate provides the oil-in-water emulsion and incorporates the active agent into the oil-in-water emulsion for delivery of the active agent to the substrate upon application of the oil-in-water emulsion to the substrate.

Controlled-release composition for topical application and a method of delivering an active agent to a substrate

A silicone adhesive composition used for adhering a device or a substance to a wet surface contains (i) a silicone resin that is cohydrolysis product of a trialkyl hydrolyzable silane and an alkyl silicate in which the cohydrolysis product contains a plurality of silicon-bonded hydroxy groups; (ii) a linear organopolysiloxane fluid containing terminal silicon-bonded hydroxy groups having a viscosity above 200,000 mm/at 25&#xb0; C.; (iii) a trialkylsiloxy terminated polyorganosiloxane fluid having a viscosity of 100,000-600,000 mm/at 25&#xb0; C.; (iv) a water soluble muco-adhesive polymer selected from the group consisting of a poly(ethylene oxide) polymer having a molecular weight of 100,000 to 8,000,000, an acrylic acid polymer having a molecular weight of 500,000 to 4,500,000,000, and a hydroxyethylcellulose polymer having a molecular weight of 90,000 to 1,300,000,000; and optionally, (v) a solvent selected from the group consisting of aliphatic hydrocarbons, aromatic hydrocarbons, esters, volatile linear and cyclic silicone compounds. Other components such as a silicone polyether, a silicone wax, a drug, an excipient, and/or an active ingredient can also be included.

Silicone adhesive for adhesion to wet surfaces

A transdermal drug delivery device for the controlled transdermal delivery of an active pharmaceutical agent. The device comprises a drug delivery device comprising an active pharmaceutical agent and a means for its controlled delivery through the skin; and (B) a silicone pressure sensitive adhesive for maintaining contact between the device and the skin of a wearer. The silicone pressure sensitive adhesive comprises silicone resin copolymer and a polydiorganosiloxane and has a silanol concentration in the range of between about 8000 and about 13,000 ppm. The present invention also relates to methods for producing the above silicone pressure sensitive adhesive compositions.

Pressure sensitive adhesive compositions for transdermal drug delivery devices

A flexible ocular insert device adapted for the controlled sustained release of an ophthalmic drug into the eye. In one embodiment, the device includes an elongated body of a polymeric material in the form of a rod or tube containing a pharmaceutically active ingredient and with at least two anchoring protrusions extending radially outwardly from the body. The device has a length of at least 8 mm and the diameter of its body portion including the protrusions does not exceed 1.9 mm. The sustained release mechanism may, for example, be by diffusion or by osmosis or bioerosion. The insert device is advantageously inserted into the upper or lower fornix of the eye so as to be independent of movement of the eye by virtue of the fornix anatomy. The protrusions may be of various shapes such as, for example, ribs, screw threads, dimples or bumps, truncated cone-shaped segments or winding braid segments. In a further embodiment, the polymeric material for the body is selected as one which swells in a liquid environment. Thus a device of smaller initial size may be employed. The present insert device is of a size and configuration such that, upon insertion into the upper or lower fornix, the device remains out of the field of vision so as to be well retained in place and imperceptible by a patient over a prolonged period of use. The device can be retained in the upper or lower fornix for 7 to 14 days or longer.

Ocular insert with anchoring protrusions

The instant invention pertains to a method of controlling release of active agents or drugs from a silicone rubber matrix by controlling the degree of crosslinking in the silicone rubber matrix. The silicone rubber matrixes of the instant invention are produced from liquid silicone rubbers comprised of (A) a polyorganosiloxane containing at least two alkenyl radicals per molecule; (B) an organohydrogensiloxane containing at least two silicon-bonded hydrogen atoms per molecule; and (C) a hydrosilylation catalyst in an amount sufficient to promote curing of said composition.
