Novel non-competitive assay techniques have been developed which not only improve sensitivity, but also are convenient and less susceptible to interfering factors. They are compatible with existing instruments and can be run in one or more test tubes. The analyte is reacted with labeled specific binder, after which the mixture is reacted with (1) an insoluble material attached to an analyte derivative and (2) a solid phase carrying a binder. The solid phase is then separated, and the label attached to the solid phase is measured. Immunoassays for measuring thyroid hormones such as thyroxine(T.sub.4) and triiodothyronine(T.sub.3) are carried out by incubating a serum sample with either (a) unlabeled thyroglobulin and a labeled antibody to a thyroid hormone or to thyroxine binding globulin(TBG), or (B) labeled thyrogloublin and an unlabeled antibody to a thyroid hormone. The thyroglobulin may be immobilized on an insoluble carrier and the immobilized thyroglobulin is preferably modified by succinylation with succinic anhydride. Whole thyroglobulin can be used or the thyroglobulin can be fragmented into smaller peptides prior to use provided the peptides contain at least one T.sub.3 or T.sub.4 residue per peptide. Preferably, the antibody is a monocional antibody and labeling is with an acridinium ester.

The present invention relates to a process for forming stable, uniform unilamellar or oligolamellar liposomes which encapsulate a macromolecule. However, unlike prior processes, the present method maintains a temperature below the phase transition temperature of the lipid used to make the liposome both during the hydration of the lipid and, if used, during an extrusion step. In addition, the liposome is formed and the macromolecule encapsulated in a low ionic strength solution, then they are dialyzed against a high ionic strength solution.
