A filament or printing material placed in a syringe for 3D printing comprising polymers, proteins, and/or functional particles and materials is provided. Methods of treating a bone defect in a subject in need thereof comprising using a handheld 3D printer to apply a filament or the printing material placed in a syringe to the bone defect of the subject are also provided. Methods of fixing or gluing natural or synthetic bone grafts using a handheld 3D printer to apply a filament or the printing material placed in a syringe over and around the defect or at the interface of a flap and the bone. Methods of printing a graft cage for retaining bone grafts and/or bone graft substitute in its desired location during healing for treatment of critical-sized segmental defects in long bones are provided.

Coaxial electrospinning is used to encapsulate a chitin-lignin based hybrid gel with polycaprolactone (PCL). Antibiotics and/or other bioactive agents loaded into the core and/or shell layer of the fibrous platform are released in a controlled and sustained manner that effectively inhibits both Gram-positive and Gram-negative bacteria without cytotoxicity to mammalian cells. The PCL shell layer provides longer life for the CL gels in a wet environment and allows sustainable drug release. The PCL-coated CL nanofiber scaffolds can be loaded with antimicrobial nanoparticles, antibiotics, oxygen-releasing agents, antioxidants and/or growth factors that promote healing when used as a controlled drug release dressing for chronic wounds, such as diabetic ulcers.

A cryogel which includes lignin nanoparticles embedded in crosslinked gelatin. The cryogel is formed at subfreezing temperatures where aggregation of polymerized or crosslinked materials is prevented by ice crystals. When thawed, the cryogel is a densely crosslinked microporous composition with a plurality of pores ranging from 50-150 &#x3bc;m in size. The cryogel combines a natural bio waste polymer in the form of lignin together with a collagen derived natural polymer. Incorporation of lignin within gelatin improves the mechanical performance of the cryogel and enhances its shape recovery rate. The macropororous composition inhibits growth of both gram positive and gram negative bacteria. In addition, the macoporous composition exhibits excellent free radical scavenging activity. All of the properties make the cryogel an excellent material for wound healing and tissue engineering applications.

Antioxidant, antibacterial, injectable lignin-gelatin composite cryogels for wound healing and tissue engineering

Inserts for the wells of tissue culture plates are provided. When placed in a tissue culture well, a bottom surface of a shaft portion of an insert contacts a bottom surface of the well and blocks the growth of cells in that area. The blocked area mimics a 'wound' in the layer of cells that grows around it. A cover portion of the insert comprises open channels through which cells, media, etc. can be delivered and gases exchanged while the insert is in place. Once the insert is removed, cell culture with the artificial 'wound' is used to test the efficacy of agents with respect to their ability to facilitate cell growth into and closure of the &#x201c;wound&#x201d;.
