Agent for preventing or ameliorating skin aging

The present invention provides a human photoaged skin model and an animal model, and a method of making the same, for evaluating cosmetics and similar products in terms of their anti-aging or rejuvenating effect.

Human photoaged skin model

A method for evaluating or selecting a substance capable of preventing or curing photodamage of skin is provided. Provided is a method for evaluating or selecting an agent for preventing or curing photodamage of skin, the method including: (A) contacting cells that are capable of expressing TIMP-1 gene or TIMP-1 protein, with a test substance; (B) measuring the expression level of the TIMP-1 gene or the TIMP-1 protein in the cells; (C) comparing the expression level obtained in (B), with the expression level of TIMP-1 gene or TIMP-1 protein in a control group in which the cells capable of expressing TIMP-1 gene or TIMP-1 protein have not been contacted with the test substance; and (D) evaluating or selecting the test substance which increases the expression level of TIMP-1 gene or TIMP-1 protein, as an agent for preventing or curing photodamage of skin, based on the results of (C).

Method for evaluating or selecting agent for preventing or curing photodamage of skin

There is provided a screening method which can select a photoaging-preventing or improving agent or a wrinkle-preventing or improving agent with efficiency and reliability. The screening method for a photoaging- or wrinkle-preventing or improving agent selects, as a candidate, a substance inhibiting the expression of keratin 6 and/or keratin 16 genes or keratin 6 and/or keratin 16 proteins in a skin tissue or cell.

Skin pore minimizers and skin elasticity improvers

The invention provides a human photoaged skin model and an animal model which are useful for evaluating cosmetics and similar products in terms of their anti-aging or rejuvenating effect, as well as a method for producing such a skin model and such an animal model. The method of producing the human photoaged skin model is characterized in that a transplanted skin area of an immunodeficient non-human animal which has undergone transplantation of human skin is irradiated with UV-B light of 80-100 mJ/cmfor consecutive 4 to 8 weeks, and the irradiated area is left to take its own course for at least 3 weeks. A human photoaged skin model and an animal model produced through this method are also disclosed.

A non-aqueous cosmetic composition contains (A) activated zeolite, (B) a polyhydric alcohol having a molecular weight not higher than 1,000, and (C) a high molecular compound soluble in the ingredient (B). When shaken upon use, the cosmetic composition permits ready and even dispersion of the zeolite, exhibits good spreadability and compatibility on the skin and can be readily applied over a wide area. Moreover, the cosmetic composition also has excellent massaging effect.

A wrinkle-care product, an aging-preventive cosmetic and a skin cosmetic each comprising a guanidine derivative of formula (I) or an acid addition salt thereof: ##STR1## wherein R1 represents a hydrogen atom, a lower alkyl group or --(AO).sub.m --(BO).sub.n --D--E �wherein A and B may be the same or different and each represents an alkylene group having 2 to 8 carbon atoms; D represents a binding hand, --CO--, or an unsubstituted or substituted alkylene group having 1 to 6 carbon atoms; E represents a hydrogen atom, a lower alkyl group, an aralkyl group or an unsubstituted or substituted aryl group; m is a number of from 1 to 6; and n is a number of from 0 to 6!; k is a number of from 1 to 10; and G represents a hydrogen atom, a hydroxyl group, a carboxyl group, a sulfonate group or a phosphate group. These products are excellent in the effects of inhibiting wrinkling and smoothing wrinkles without damaging the physiological conditions of the skin.

A composition for external skin care comprising an amine derivative represented by the following formula (1), ##STR1## wherein R.sup.1 is a linear, branched, or cyclic saturated or unsaturated hydrocarbon group having 4-40 carbon atoms, and R.sup.2, R.sup.3, R.sup.4, R.sup.5, and R.sup.6 are individually a hydrogen atom or a hydrocarbon group having 1-10 carbon atoms, which may be substituted or unsubstituted by 1 or more hydroxy groups; or an acid addition salt thereof, in an amount from 0.0001 to less than 0.1% by weight; and a method of curing wrinkles and keratinization of the skin characterized by applying said amine derivative or its salt to the skin.
