An assay chip includes fluidic-channel member composed of a light-transmissive lower member and an upper member, forming a fluidic-channel therebetween, and a cover member fitted with the fluidic-channel member from the upper-member-side thereof. An inlet for injecting a sample solution into the fluidic-channel and a suction opening for sucking, from the downstream side, the injected sample solution, both communicating with the fluidic-channel, are formed on the upper surface of the upper member. A pot for carrying out predetermined pre-processing on the sample solution, a pot for first-reaction processing to bind a photoresponsive labeling substance to an analyte in the sample solution, an inlet insertion-hole for inserting the inlet, and a suction-opening insertion-hole for inserting the suction opening are linearly arranged on the upper surface of the cover member.

An object to be achieved by the present invention is to provide a nucleic acid amplification method by which a nucleic acid can be amplified substantially isothermally using oligonucleotide primers and DNA polymerase capable of strand displacement. The present invention provides a nucleic acid amplification method which comprises performing substantially isothermal incubation of a reaction solution containing at least one type of deoxynucleotide triphosphate, at least one type of DNA polymerase having strand displacement activity, a divalent cation, at least 0.01% or more surfactant, at least two types of oligonucleotide primer, and the nucleic acid fragment as a template so as to perform a polymerase reaction that initiates from the 3' end of the primer and thus amplifying the nucleic acid fragment.

Isothermal nucleic acid amplification method using surfactant

An object to be achieved by the present invention is to provide a nucleic acid amplification method by which a nucleic acid can be amplified using oligonucleotide primers and DNA polymerase. The present invention provides a nucleic acid amplification method which comprises performing incubation of a reaction solution containing at least one type of deoxynucleotide triphosphate, at least one type of DNA polymerase, at least two types of oligonucleotide primer, and the nucleic acid fragment as a template so as to perform a polymerase reaction that initiates from the 3' end of the primer and thus amplifying the nucleic acid fragment, wherein a tag sequence is added at the 5&#x2032; end of the first oligonucleotide primer, and the tag sequence is a nucleotide sequence on the template nucleic acid fragment which is present downstream of the sequence which is substantially complementary with the 3&#x2032; end region of the first oligonucleotide primer (a region where the first oligonucleotide is annealed to the template nucleic acid).

The present invention describes methods for discriminating between nucleotide sequences of first and second nucleic acids, including: providing a reaction solution, including a deoxynucleotide triphosphate, a DNA polymerase with strand displacement ability, a template nucleic acid fragment, a primer, and a mask oligo; incubating the reaction solution, obtaining a polymerase reaction, and producing an amplification product; detecting the amplification product to discriminate between the nucleotide sequences, wherein the primer is complementary to the first nucleic acid, the mask oligo hybridizes to the nucleotide sequence portion of the first and second nucleic acid, wherein the mask oligo is more complementary to the second nucleic acid than to the first nucleic acid, and wherein the mask oligo is not an origin of an elongation reaction with the polymerase, and a primer portion and a mask oligo portion hybridize to the same regions on the first and second nucleic acid.

Method for discriminating between nucleotide sequences of nucleic acids

The immunoassay element for quantitatively analyzing an antigen by determining the change in enzymatic activity of an enzyme-labelled antigen or antibody caused by an immunological reaction. The immunoassay element comprises a substrate layer containing a non-diffusible substrate which forms a diffusible material in the presence of the labelling enzyme, and a reagent layer containing a fragmenting enzyme for further fragmenting the diffusible material into a lower molecular weight product. As the non-diffusible substrate, a substrate capable of reacting solely with the labelling enzyme and incapable of reacting the fragmenting enzyme is utilized. When an endo-active glucosidase is used as the labelling enzyme, and an exo-active glucosidase is used the fragmenting enzyme in the reagent layer, the non-diffusible substrate of the substrate layer is preferred to be an endo type selectively reactive substrate, which means a substrate having a reactivity specific to endo-active glucosidase. Highly sensitive assay is realized with high accuracy and high reproducibility and good storage stability.

A method of automatically isolating and purifying nucleic acid from a nucleic acid-containing specimen is provided, the method comprising: injecting a liquid into a cartridge for isolation and purification of a nucleic acid including at least two openings from one opening of the at least two openings, in which the cartridge includes a container having the at least two openings and containing a nucleic acid-adsorbent solid phase; passing the liquid through the nucleic acid-adsorbent solid phase by a pressure difference generated by a pressure generation means for generating a pressure difference between the inside and outside of the container; and discharging the liquid from the other opening of the container to the outside of the container by a pressure difference generated by the pressure generation means, wherein a pressure generated in the inside of the container by the pressure generation means is measured, a pressure change velocity and a pressure change acceleration are calculated on the basis of the value of the measured pressure, and the timing of completion of discharge of the liquid from the container is determined by use of a temporal change pattern of at least one of the measured pressure, the pressure change velocity and the pressure change acceleration.

In an ink jet recording head including a flexible wiring board, the flexible wiring board has a thickness reduction part formed by reducing a film thickness of a part of the base film. The thickness reduction part is configured to alleviate stress applied to inner leads by a base film in the periphery of a device hole, so as to avoid the failure of bonding or the like caused by the inner leads of the flexible wiring board, on which a recording element substrate is installed.

The present invention is a method for isolating and purifying a nucleic acid, where generation of foams is able to be suppressed whereby the isolation and purification of a nucleic acid are easily and efficiently carried out, the method for isolating and purifying a nucleic acid comprising the step of: (1) contacting a sample solution containing nucleic acid to a solid phase to adsorb the nucleic acid onto the solid phase; (2) contacting a washing solution to the solid phase to wash the solid phase in such a state that the nucleic acid is adsorbed; and (3) contacting an elution solution to the solid phase to desorb the nucleic acid, wherein the sample solution containing nucleic acid contains an antifoaming agent.
