Disclosed is a measurement apparatus for analyzing a cell contained in a specimen, comprising: a chamber for preparing a measurement sample in which the cell is stained with first and second fluorescent dyes contained in a reagent supplied from at least one reagent container; a liquid feeding section for feeding the reagent from the reagent container to the chamber via a liquid feeding tube provided between the reagent container and the chamber; and a detection section that acquires first and second signals each corresponding to fluorescence of a first wavelength and fluorescence of a second wavelength emitted from the cell stained with the first and second fluorescent dyes in response to irradiation of the measurement sample flowing in a flow cell with light; and an analysis section that analyzes the cell on the basis of the first and second signals.

The present invention provides a method for classifying and counting leukocytes which allows classification and count of normal leukocytes as well as discrimination between blast cells and atypical lymphocytes. A method for counting megakaryocytes in a specimen is described. In the method, first, erythrocytes in the specimen are lysed and nucleic acid in the megakaryocytes is stained with a fluorescent dye, and thereby, a measurement sample is prepared. Next, the cells in the measurement sample are irradiated with excited light so that the forward scattered light, the side scattered light and the fluorescence, which are emitted from the cells, are detected. Megakaryocytes are identified on the basis of the detected forward scattered light, the fluorescence and the side scattered light. Then, the identified megakaryocytes are counted.

Reagent and method for classification and counting of leukocytes

A hematological sample is treated with a hemolytic agent that maintains immature leukocytes in a viable state and damages other leukocytes. The damaged leukocytes are stained with a fluorochrome which can stain damaged cells. At least one kind of scattered light and at least one kind of fluorescence of the treated and stained blood cells are measured to classify and count leukocytes based on the intensities of the scattered light and the fluorescence. Highly precise measurement of immature leukocytes and simultaneous classification and counting of normal leukocytes are achieved.
