This disclosure concerns the use of endogenous plant RNAi machinery to preferentially or specifically reduce transgene expression. In some embodiments, the disclosure concerns specific reduction of transgene expression in seed tissues of a dicot plant.

The subject invention relates in part to the surprising discovery that CryDa is active against corn earworm (CEW),(Boddie). Methods for using CryDa in transgenic plants to prevent serious crop damage is described. Leaf and silk bioassays using transgenic maize expressing full length, core toxin region or chimeric CryDa demonstrated good insect protection against CEW larvae damage.

Described herein are murine monoclonal antibodies and methods useful for determining and quantitating the presence of Cry1Ca delta endotoxin. The claimed antibodies specifically bind the core toxin region making them suitable for detecting the native full length Cry1Ca toxin as well as the amino core toxin and N-terminal 29 residue truncated forms.

The present invention provides methods, vectors and gene constructs for enhancing expression of a recombinant nucleic acid sequence in transgenic plants and plant tissues. According to the present invention, nucleic acid sequences are obtained and/or derived from the 5' and 3&#x2032; untranslated regions of genes encoding osmotin proteins and engineered to flank respective portions of a selected coding region of a vector. The vector construct may be introduced into plants and/or plant tissues through conventional procedures, resulting in enhanced expression of the selected coding region.
