A bubble point of a combination of a filter and a liquid is calibrated by applying an initial back pressure through the filter toward the liquid; taking a first plurality of measurements of the level of the liquid in the container; calculating a first variance of the first plurality of measurements; and comparing the first calculated variance with a known threshold variance. Then a second plurality of measurements of the level of the liquid in the container is taken; a second variance of the second plurality of measurements is calculated; and the second calculated variance is compared with a known threshold variance. The above described steps are repeated at incrementally increased back pressures until the first and second calculated variances are each greater than or equal to the known threshold variance, which is about 0.01. The bubble point is determined to be the back pressure at that point. If the determined bubble point is less than or equal to the known threshold bubble point, which is about 0.8 pounds per square inch, the filter is identified as unsuitable for use in cell block formation.

Method and apparatus for calibrating filters during specimen collection

A system for making cell blocks includes a cell block cassette and processing station, the cassette including a main body having a having a collection aperture formed therein and a filter assembly removably attached to the main body, the filter assembly defining a collection well in communication with the collection aperture, and having a filter positioned across a bottom surface of the collection well, the filter configured to retain cellular matter carried in a fluid that is dispensed into the collection well and flows across the filter. The cell block processor has a cassette interface removably seating the cell block cassette, and a sensor positioned or positionable to detect and monitor a fluid level in the collection well. The processing station includes an automated fluid delivery system operable to dispense a fluid into the collection well, and a controller operatively coupled to the fluid delivery system, wherein the controller causes the fluid delivery system to selectively dispense fluids into the collection well based at least in part on a flow rate across the filter determined at least in part based on changes in the monitored fluid level in the collection well.

Methods for processing cell block

In various aspects, the present teaching provide systems for providing sample ions, methods for providing sample ions, and sample support handling mechanisms. The sample support handling mechanisms of the present teachings comprising a sample support transfer mechanism portion and a sample support changing mechanism portion, where the sample support changing mechanism portion is disposed in a vacuum lock chamber. In various embodiments, the sample support handling mechanism facilitates providing consistent positioning of a sample support for subsequent ion generation by MALDI.

Sample handling mechanisms and methods for mass spectrometry

A MALDI plate construction is provided to support a sample in a manner to effect mass spectrometry of the sample. The plate construction comprises a sample receiving surface, a plate holder for retaining the sample receiving surface and an electrically conductive grid positioned adjacent to the sample receiving surface and in electrical contact with the plate holder.

A sample plate structure is provided including a retainer plate having a central opening, a sample insert plate which fits into the opening, an optional pressure plate and a handle. The handle applies pressure to the sample insert plate to retain it within the retainer plate. The sample insert plate is provided with a peripheral configuration, which assures that the sample insert plate is properly oriented within the sample plate support structure and is held flat. While the sample insert plate can be a consumable, the remaining portion of the apparatus can be reused.

A sample plate structure is provided including a retainer plate having a central recess with a trough along its periphery, a sample insert plate which fits into and rests on contact surface of the recess, and a magnet that is held below the contact surface. A portion of the bottom surface of the insert is formed of a magnetic material. The magnet provides sufficient force to retain the insert plate in the retainer plate during MALDI MS analysis. The sample insert plate is provided with a peripheral configuration, which assures that the sample insert plate is properly oriented within the sample plate support structure and is held flat. A hole for a protrusion allows easy insert installation and alignment against the orientation feature in the recess as well as easy removal of the insert simply by pushing up from underneath the retainer plate. While the insert sample plate can be a consumable, the remaining portion of the apparatus can be reused.
