The present invention relates to crystal forms of a replication protein encoded by a plasmid isolated from a Multiple Sclerosis patient, crystal structure information obtained from them, methods of preparing such crystal forms, their use for the identification and/or design of inhibitors of said replication proteins and methods for identifying, optimizing and designing compounds which should have the ability to interact with or inhibit the replication protein.

Disclosed is a method of diagnosing multiple sclerosis (MS), wherein a blood sample from a patient is incubated with a DNA-replication associated (REP) protein. The present invention relates to a DNA-replication-associated (Rep) protein for use in the diagnosis of multiple sclerosis (MS), wherein (a) an increased amount of Rep protein or fragments thereof in the sample as compared to an amount in a control sample; or an increased amount of anti-Rep protein antibodies with antigen in a sample from a subject as compared to an amount in a control sample correlates with a diagnosis of MS, wherein the Rep protein is MSBI1 Rep or MSBI2 Rep.

The present invention concerns a DNA-replication-associated (Rep) protein comprising an amino acid sequence as depicted in SEQ ID NO: 11 or 12; (b) a fragment of SEQ ID NOs:11 or 12 which is capable of binding an anti-Rep antibody specific for a protein having the amino acid sequence of SEQ ID NOs: 11 or 12; or (c) an amino acid sequence having a 90% or more homology to the amino acid sequence of (a) or (b) and is capable of binding an anti-Rep antibody specific for a protein having an amino acid sequence of SEQ ID NOs:11 or 12. The present invention further concerns a method of diagnosing MS or a predisposition for MS and a kit for use in such methods.
