Method and device for purifying nucleic acids

A process and closure arrangement is provided for isolating purified biological material. The process includes placing biological materials in separate lysis vessels, adding lysis liquids to the biological materials in the lysis vessels, and transferring the liquids in the lysis vessels into a matrix unit. The liquids in the matrix vessels of the matrix unit are extracted through outlet openings during which the liquids flow through matrices and the matrix unit is placed on a collecting unit with collecting vessels which are arranged such that at least the outlet openings of the matrix vessels extend into the collecting vessels. The matrix vessels are with elution fluid and elution fluids from the matrix vessels are extracted through their outlet openings during which the elution fluids flow through the matrices and the elution fluids which are enriched with biological material are collected in the collecting vessels.

System for providing biological materials

The present invention is directed to a method for adsorbing, i.e. non-covalently binding, nucleic acids to a solid phase using a two-step procedure. Furthermore, the present invention pertains to a method for isolating nucleic acids from a biological sample. In the first step of the procedure, lysis is effected by mixing the biological sample with an aqueous lysis buffer containing a chaotropic agent and incubating the mixture; in the second step, the concentration of the chaotropic agent in the mixture is increased and the mixture is contacted with the solid phase, whereby the nucleic acids in the liquid phase is adsorbed to the solid phase.

Adsorption of nucleic acids to a solid phase

Magnetic particles are prepared containing a magnetic core coated with a glass layer having a substantially pore-free glass surface or having pores with a diameter of less than 10 nm. The particles are used for separating biological material such as nucleic acids. A preferred process of preparing the particles is by forming a mixture of magnetic cores with a sol formed from an alcohol and a metal alkoxide, spray-drying the mixture to coat the cores with a layer of gelled sol, and heating the coated cores to obtain the magnetic glass particles. Preferably, the particles have an average particle size of less than 100 &#x3bc;m. The magnetic core may be a composite material containing a mica core and magnetite particles immobilized on the mica core, and the glass layer may contain boron oxide. Magnetic core materials include magnetite (FeO) and FeO.

Method for separating biological material from a fluid using magnetic particles

A method for purification of a nucleic acid comprising the steps of (a) adsorbing on a substrate the nucleic acid from a composition containing (i) an aqueous buffer, (ii) salts in a high concentration, (iii) a water-miscible, non-acidic organic compound, and (iv) the nucleic acid; (b) optionally washing with a washing solution the substrate with the adsorbed nucleic acid; (c) contacting the substrate with the adsorbed nucleic acid with a solution containing salts in a lower concentration compared to the composition of step (a), thereby desorbing the nucleic acid from the substrate; (d) separating the solution with the desorbed nucleic acid from the substrate, thereby purifying the nucleic acid; and optionally (e) precipitating the desorbed nucleic acid from the solution of step (d) and isolating the precipitated nucleic acid, thereby further purifying the nucleic acid.

A device for isolating nucleic acids which reduces the transfer of contaminants during nucleic acid isolation methods which is composed of two vessels which are linked by a closing element in which a material that binds nucleic acids is placed. The nucleic acid can for example be bound to the material by tipping the device.

In using the magnetic glass particles to separate a biological material, the particles are contacted with a fluid containing the biological material such that the biological material binds to the glass surface, and the bound biological material is separated from the fluid such as by using a magnetic field. Before applying a magnetic field, the magnetic particles may sediment when contacted with the biological material.

Particles having a magnetic core and outer glass layer for separating biological material

Device for the isolation of nucleic acids from a liquid sample by introducing the liquid into the device through an inlet opening and passage of the sample through a nucleic acid binding material and discharging of the resulting liquid through an outlet opening, wherein the nucleic acid binding material is inserted through the outlet opening during the manufacture of the device and the outlet opening is narrowed in order to hold the material. The device is particularly simple and reliable to manufacture but yet ensures that nucleic acids can be isolated without contamination.
