The invention provides lipid bodies isolated from yeast, compositions comprising the lipid bodies, products made from the lipid bodies, methods of making the lipid bodies, and methods of using the lipid bodies. The lipid bodies of the invention have an exceptionally large size and high internal neutral lipid content, providing a number of advantages for a variety of practical applications.

Lipogenic yeasts bioengineered to overexpress genes for lipid production, and methods of use thereof. The yeasts are modified to express, constitutively express, or overexpress an acetyl-CoA carboxylase, an alpha-amylase, an ATP citrate lyase, a diacylglycerol acyltransferase, a fatty acid synthase, a glycerol kinase, a 6-phosphogluconate dehydrogenase, a glycerol-3-phosphate dehydrogenase, a malic enzyme, a fatty acyl-CoA reductase, a delta-9 acyl-CoA desaturase, a glycerol-3-phosphate acyltransferase, a lysophosphatidate acyltransferase, a glucose-6-phosphate dehydrogenase, a beta-glucosidase, a hexose transporter, a glycerol transporter, a glycoside hydrolase enzyme, an auxiliary activity family 9 enzyme, or combinations thereof. The yeasts in some cases are also modified to reduce or ablate activity of certain proteins. The methods include cultivating the yeast to convert low value soluble organic stillage byproducts into lipids suitable for biodiesel production and other higher value uses.

Disclosed are nucleic acid constructs comprising coding sequences operably linked to a promoter not natively associated with the coding sequence. The coding sequences encodeproteins that allow recombinant strains ofexpressing the protein to grow on xylose, and allow or increase uptake of xylose byorexpressing the coding sequences. Expression of the coding sequences enhances uptake of xylose and/or glucose, allowing increased ethanol or xylitol production.

Also disclosed are substantially purified preparations of xylanase enzymes from bacterial isolates. The xylanase is preferrably isolated xyl 3 obtained from Streptomyces roseiscleroticus NRRL B-11019, wherein xyl 3 has a molecular weight of approximately 21 kD as determined by SDS-gel electrophoresis and a pH optima of pH 5.0-7.0.

A method of removing color from wood pulp is disclosed. The method comprises the steps of preparing a wood pulp, treating the wood pulp with the xylanase wherein the xylanase is capable of releasing chromophores from the pulp and extracting the wood pulp to remove chromophores. In a preferred form of the invention, the wood pulp is a kraft pulp and the xylanase is selected from the group consisting of xyl 1, xyl 2, xyl 3 and xyl 4. These xylanases are obtained from Streptomyces roseiscleroticus NRRL-11019.
