A skull-mounted drug and pressure sensor (SOS), a smart pump (ISP) electrically coupled to the SOS and a drug delivery and communications catheter communicating the SOS with the ISP are combined for a first embodiment. A skull-mounted (SOS), a metronomic biofeedback pump (MBP) electrically coupled to the SOS and a drug delivery and communications catheter having a sending and receiving optical fiber communicating the SOS with the MBP are combined for a second embodiment. A third embodiment combines a (SOS), an implantable power and communication unit (PCU) electrically coupled to the SOS, and a drug delivery and communications catheter for communicating the SOS with the PCU and for communicating the exterior source of the drug to the SOS. A fourth embodiment combines a ventricular catheter with a CSF accessible chamber and drug delivery port; and an implantable stand-alone skull-mounted drug and pressure sensor (SPS).

Colloidal suspensions comprising a therapeutic agent and squalene. Squalene binds to certain compounds, such as gacyclidine, much more tightly than other drug carriers, such as polylactic glycolic acid. Including squalene in the particulate phase sequesters the therapeutic agent and provides superior stability at room or body temperature.

Drug solutions (or other combinations of vehicle with entrained drug) are prepared by removing drug from one or more masses of a solid form of the drug. The solid form of the drug may be sparingly soluble or insoluble in water. Examples of devices for holding solid drug and facilitating delivery of such drug to targeted regions are also described.

Flow-induced delivery from a drug mass

Implantable drug delivery systems target delivery of small volumes of drugs to specific tissues. In some cases, a drug delivery system includes an implantable osmotic pump connected to a drug-containing housing, with that housing connected to a needle, cochlear implant or other type of component for ultimate delivery to the target tissue. In some implementations, a subcutaneous port receives a fluid from an external pump. The port is connected to a needle or other component for delivery of one or more drugs to the target tissue. Both solid and liquid drug formulations can be used. In embodiments using solid drugs, a separate drug vehicle (such as saline) can be used to dissolve a portion of the solid drug, with the drug-loaded vehicle then delivered to the target tissue.

Apparatus and method for delivery of therapeutic and other types of agents

The prodrug of the invention is a modified form of a therapeutic agent and comprises a therapeutic agent, an oligopeptide, a stabilizing group and, optionally, a linker group. The prodrug is cleavable by the enzyme Thimet oligopeptidase, or TOP. Also disclosed are methods of designing prodrugs by utilizing TOP-cleavable sequences within the conjugate and methods of treating patients with prodrugs of the invention.

Enzyme-cleavable prodrug compounds

The present invention provides analogues of duocarmycins that are potent cytotoxins. Also provided are peptidyl and disulfide linkers that are cleaved in vivo. The linkers are of use in forming prodrugs and conjugates of the cytotoxins of the invention as well as other diagnostic and therapeutic moieties. The invention provides prodrugs and conjugates of the duocarmycin analogues with the linker arms of the invention.

Methods of treating cancer with an antibody-drug conjugate

The prodrug of the invention is a modified form of a therapeutic agent and comprises a therapeutic agent, an oligopeptide of three amino acids, a stabilizing group and, optionally, a linker group. The prodrug is cleavable by a trouase enzyme such as Thimet oligopeptidase. Also disclosed are methods of making and using the prodrug compounds.

Tripeptide prodrug compounds

The compounds of the invention are modified forms of therapeutic agents. A typical prodrug compound of the invention comprises a therapeutic agent, an oligopeptide having an isoleucine residue, a stabilizing group and, optionally, a linker group. The prodrug is cleavable by an enzyme associated with the target cell. Methods of making and using the compounds are also disclosed.
