Assay methods utilizing the response of a magnetically responsive reagent to the influence of a magnetic field to qualitatively or quantitatively measure binding between specific binding pair members. According to the invention, the presence of an analyte mediates whether or not the magnetically responsive reagent binds to a mobile solid phase reagent. The extent of binding will modulate the response of the magnetically responsive reagent or that of the mobile solid phase reagent, or both, to the influence of a magnetic field. Hence, by measuring the response to the magnetic field of the magnetically responsive reagent, or that of the mobile solid phase reagent, the presence or amount of analyte contained in a test sample can accurately be determined. The invention utilizes various devices to carry out the assay methods described.

A method, composition, device, apparatus, and kit for the determination of the presence or amount of an analyte by monitoring an analyte-mediated ligand binding event in a test mixture which contains the analyte to be assayed, a specific binding member, a Raman-active label, and a particulate having a surface capable of inducing a surface-enhanced Raman light scattering. The test mixture is illuminated with a radiation sufficient to cause the Raman-active label in the test mixture to emit a detectable Raman spectrum. The present invention provides assay methods for performing binding assays, wherein the detectable label is a magnetically responsive material. Direct and indirect, competitive and sandwich assay formats are used to partition the magnetically attractable label between a solid phase and a fluid phase in proportion to the presence or amount of analyte in the test sample. The magnetic responsiveness of the magnetically attractable label in one or both phases results in the exertion of a force upon the label. By determining the extent of the force or influence of the force exerted upon the label, the amount of the analyte in the test sample is determined.

The present invention provides devices for performing binding assays. An assay for an analyte in a test sample that uses a ligand binding reaction between the analyte and a specific binding member or a binding member pair that includes one binding member having a Raman active reporter. The analyte can be assayed by measurement of the Raman scattering signal of the Raman-active reporter. The Raman scattering signal can be either a noon-enhanced Raman scattering signal or a Raman scattering signal that is enhanced by a surface capable of enhancing that Raman scattering signal.
