The present application relates to bioactive implants, methods of making bioactive implants, and methods of using bioactive implants to treat or repair a bone defect or a cartilage defect. In an aspect, the present application relates to compositions comprising bone microparticles in a solution, wherein the compositions harden upon desiccation into bioactive implants. In an aspect, the present application relates to compositions comprising cartilage microparticles in a solution, wherein the compositions harden upon desiccation into bioactive implants. In an aspect, disclosed herein are methods of making and using the disclosed compositions comprising bone microparticles and the disclosed composition comprising cartilage microparticles. This abstract is intended as a scanning tool for purposes of searching in the particular art and is not intended to be limiting of the present invention.

Bioactive implants and methods of making and using

A collagen-bone composition may include bone collagen and bone particles. The collagen may be in the form of a collagen suspension and the bone particles may be demineralized bone particles retained in collagen suspension. The composition may be prepared from pressure and thermal treatment of dry demineralized bone particles contacted with an aqueous medium to produce a viscous gel-like fluid. The fluid may include the collagen extracted from the demineralized bone particles. Addition of particulate bone may be used to increase the consistency of the composition, which may convert it into a paste or putty. The paste or putty may be suitable for ejection from a syringe. The collagen-bone composition may be preserved by freeze-drying or preferably by hypothermic dehydration. On reconstitution with aqueous medium, the preserved material may resume its original shape and properties.

Bone putty and gel systems and methods

A particulate composition for stimulating nucleus pulposus regeneration has a particulate composition made of vertebral end-plates having an osseous component wherein the composition is milled, ground or particulized into particles from 10 to 1000 microns in size. At least a part of the composition can be non-demineralized or demineralized or a mixture of demineralized and non-demineralized particles from the vertebral end-plates. Preferably, the non-demineralized part is not subjected to harsh chemical treating. To possibly enhance the release of growth factors and other similar substances from the osseous layer of the end-plate, the material may be treated with hydrochloric acid, ethylene diamine or other demineralizing agents or regimens.

The invention is a novel method of sterilizing bone and soft tissues intended for human transplantation. In one embodiment the process includes steps of subjecting transplants to high hydrostatic pressures of aqueous fluid to which chlorhexidine gluconate or similar sterilizing chemicals have been added.

An allogeneic or xenogeneic implant for intervertebral disc replacement includes a body. The body may include one or both of a plurality of grooves or tubular apertures. The body may be formed of partially decalcified bone. The plurality of tubular apertures may be defined within the body and be positioned to allow inflow of decalcifying solutions for rapid and uniform decalcification. After partial decalcification, the body may be pliable and compressible.

Modified, pliable, and compressible cortical bone for spinal fusions and other skeletal transplants

A bone plate assembly has the screws on one plate, either distal or proximal to the fracture, placed through round holes perpendicular to long axis of a first bone thus securing the plate rigidly to the bone on one side relative to the fracture. The screw holes can be either threaded on non-threaded. On the other side of the fracture, the bone fragment is first secured with an inclined screw placed from less than 90 to 10 degrees towards the fracture. As the screw is tightened it advances away from the fracture pulling the first bone, securely attached, plate towards the fracture site and thus compressing the fracture.

A method of preparing a bone graft of compressed dehydrated decalcified trabecular bone may include compressing the bone while hydrated from a first form to a second form such that the bone in the second form is compressed relative to the first form. The bone may be dried while compressed. When dry, the bone substantially retains the second form and does not revert to the first form when the compression is removed.

Cartilage materials such as cartilage fluff and a cartilage composition comprising a particulate material are disclosed. These are suitable for stimulating chondrogenesis and/or producing cartilage regeneration. Also disclosed are processes for their preparation. Methods for regenerating articular cartilage are also disclosed, which involve, for example, placing the cartilage fluff or cartilage composition into a cartilage defect.

The spinal cord tissue is dried or has the water content in the tissue reduced or eliminated by freeze drying or hypothermic dehydration and treated with a cryopreservative.

The thick fan-folded grafts of fascia lata cruciate ligament substitutes disclosed herein are used in a single-loop fashion and have surprisingly exhibited load to failure and stiffness values that compare favorably with conventional grafts, including tibialis anterior, tibialis posterior, peroneus longus, and bone-patellar tendon-bone (BPTB) half-patella specimens, when the harvested iliotibial band grafts of fascia lata avoid portions thicker than 5 mm, or even 3 mm.

Fan folded fascia lata for cruciate ligament substitution and method and apparatus for making the same

A flexible, bendable organic decalcified or partially decalcified bone, cortical or cancellous, adapted for use in augmentation or repair of animal skeletal structures comprising a continuous plate or sheet of natural bone, as well as dermis is described. The thickness, flexibility and tensile strength of the construct is such as to allow it to be shaped and contoured without damage to it. The composition is ultimately remodeled by the body, thus obviating the need for additional surgical intervention. The clinical indications for the use of the invented construct are many, but are particularly prominent in dentistry, oral and maxillofacial surgery and implantology. It is particularly useful in the maxillary sinus augmentation. A unique new method, different from previously described methods for the preparation of the disclosed constructs, is described.
