A micro-electrical-mechanical system (MEMS) resonator device includes at least one functionalization material arranged over at least a central portion, but less than an entirety, of a top side electrode. For an active region exhibiting greatest sensitivity at a center point and reduced sensitivity along its periphery, omitting functionalization material over at least one peripheral portion of a resonator active region prevents analyte binding in regions of lowest sensitivity. The at least one functionalization material extends a maximum length in a range of from about 20% to about 95% of an active area length and extends a maximum width in a range of from about 50% to 100% of an active area width. Methods for fabricating MEMS resonator devices are also provided.

Acoustic resonator device with controlled placement of functionalization material

The present invention relates to encapsulated microfluidic packages and methods thereof. In particular embodiments, the package includes a device, a cradle configured to support the device, and a lid having a bonding surface configured to provide a fluidic seal between itself and the device and/or cradle. Other package configurations, as well as methods for making such fluidic seals, are described herein.

Microfluidic package and method of making the same

Viruses and other bioagents are of high medical and biodefense concern and their detection at concentrations well below the threshold necessary to cause health hazards continues to be a challenge with respect to sensitivity, specificity, and selectivity. Ideally, assays for accurate and real time detection of viral agents and other bioagents would not necessitate any pre-processing of the analyte, which would make them applicable for example to bodily fluids (blood, sputum) and man-made as well as naturally occurring bodies of water (pools, rivers). We describe herein a robust biosensor that combines the sensitivity of surface acoustic waves (SAW) generated at a frequency of 325 MHz with the specificity provided by antibodies and other ligands for the detection of viral agents. In preferred embodiments, a lithium tantalate based SAW transducer with silicon dioxide waveguide sensor platform featuring three test and one reference delay lines was used to adsorb antibodies directed against Coxsackie virus B4 or the negative-stranded category A bioagent Sin Nombre virus (SNV), a member of the genus Hantavirus, family Bunyaviridae, negative-stranded RNA viruses. Rapid detection (within seconds) of increasing concentrations of viral particles was linear over a range of order of magnitude for both viruses, although the sensor was approximately 50&#xd7;10-fold more sensitive for the detection of SNV. For both pathogens, the sensor's selectivity for its target was not compromised by the presence of confounding Herpes Simplex virus type 1. The biosensor was able to detect SNV at doses lower than the load of virus typically found in a human patient suffering from hantavirus cardiopulmonary syndrome (HCPS). Further, in a proof-of-principle real world application, the SAW biosensor was capable of selectively detecting SNV agents in complex solutions, such as naturally occurring bodies of water (river, sewage effluent) without analyte pre-processing.

Detection of bioagents using a shear horizontal surface acoustic wave biosensor

A cartridge for sample handling and sensing includes (i) a sample port; (ii) a first fluid port connected to the sample reservoir in the distal region via a first fluid channel; and (iii) a second fluid port connected to the sample reservoir via a second fluid channel. The cartridge includes (i) a sensor platform comprising a bulk acoustic wave (BAW) resonator and a fluid flow path comprising a sensing region extending across a sensing surface of the BAW resonator; and (ii) a fluid valve between the sample reservoir and the sensing region. A sample may be applied to the sample port; first volume of fluid may be injected through the first fluid port; and then a second volume of fluid may be injected through the second fluid port to drive the sample into the sensing region of the fluid flow path.

Cartridges for integrated BAW biosensors and methods for using the same

A sensor using electrophoresis may include a microfluidic channel and electrodes on opposite sides of the microfluidic channel to generate an electric field across, or normal to, the channel. The electric field may be used to drive charged particles of material, particularly material suspended in fluid in the microfluidic channel, toward or away from the one of the electrodes. The electric field may be modulated to allow material to continue flowing through the microfluidic channel, to remove non-target material, or to measure another target material.

Microfluidic sensors using electrophoresis

The present application relates to a biosensor that employs an acoustic cavity to store mechanical energy. In particular examples, the biosensor includes an electrode region and one or more reflector regions to form the acoustic cavity, as well as a functionalized active area disposed in proximity to the cavity. Methods of making and using such biosensors are also described herein.

Shear horizontal surface acoustic wave (SH-SAW) resonators and arrays thereof

Methods, structures, and systems are disclosed for biosensing and drug delivery techniques. In one aspect, a device for detecting an analyte and/or releasing a biochemical into a biological fluid can include an array of hollowed needles, in which each needle includes a protruded needle structure including an exterior wall forming a hollow interior and an opening at a terminal end of the protruded needle structure that exposes the hollow interior, and a probe inside the exterior wall to interact with one or more chemical or biological substances that come in contact with the probe via the opening to produce a probe sensing signal, and an array of wires that are coupled to probes of the array of hollowed needles, respectively, each wire being electrically conductive to transmit the probe sensing signal produced by a respective probe.

Microneedle arrays for biosensing and drug delivery

The present invention, in part, relates to methods and apparatuses for on-chip amplification and/or detection of various targets, including biological targets and any amplifiable targets. In some examples, the microculture apparatus includes a single-use, normally-closed fluidic valve that is initially maintained in the closed position by a valve element bonded to an adhesive coating. The valve is opened using a magnetic force. The valve element includes a magnetic material or metal. Such apparatuses and methods are useful for in-field or real-time detection of targets, especially in limited resource settings.

Amplification of biological targets via on-chip culture for biosensing

The present invention relates to adhesive-based microvalves, as well as assemblies and cartridges having such microvalves. In particular examples, substantially planar layers of adhesive materials are used to construct both normally-open and normally-closed microvalves for fluid control within microfluidic laminate cartridges. Such microvalves and cartridges can be useful for medical, chemical, and biological investigations.

High-temperature, adhesive-based microvalves and uses thereof

The present invention is directed to devices, systems, and methods for detecting and/or monitoring one or more markers in a sample. In particular, such devices integrate a plurality of hollow needles configured to extract or obtain a fluid sample from a subject, as well as transducers to detect a marker of interest.
